The plasmid is obtained by initially cultivating vitamin B12 non-demandable Bacillus methanolicus PB 1 (NCIMB 13113) in a methanol minimal medium at 50 deg.C overnight followed by centrifugal separation to harvest, suspending it into a buffer solution, strongly stirring the suspension after adding lysozyme, extracting with phenol in the presence of an alkali and finally agarose-electrophoresing the extract to separate the objective plasmid.

The strain is cultured in a medium produced by adding pyruvic acid to the above ribose medium, etc., and the proliferated strain is selected to obtain a pyruvic acid kinase-insufficient or deficient mutant.
