摘要： PURPOSE: To obtain an immunoconjugate having an increased selectivity to a target cell and a lowered toxicity to a receptor by reacting a protein made as a derivative by a reaction with a hetero bifunctional reagent, with an antibody or a fragment of the antibody for linking the same through a stable thioether bond. CONSTITUTION: This immunoconjugate bonding a protein with an antibody by a thioether bond, is obtained by reacting a protein (e.g.; a holotoxin, a protein for a medicinal carrier and a biological response modifier) with a compound of formula I R1 is (CH2 )n , formula II or formula III [(n) is 1-10; R2 is o- or p-nitrophenol, cyanomethyl, 2-mercaptopyrridyl, etc.]}, separating a protein derivative, reacting the protein derivative with the antibody or a fragment of the antibody under a condition capable of reducing an original disulfide bond in the antibody and forming thiol group capable of forming a thioether bond with the maleimide group of the protein derivative.

摘要： PURPOSE: To determine the reinforcement time of effector cell in an optimum organism regarding ADCC response by determining the basic level and NK or LAK cell activity of an antibody-dependent cell-mediated cytotoxicity(ADCC) using a basic effector cell sample from a patient. CONSTITUTION: The basic level of ADCC and NK or LAK cell activity are determined by a basic effector cell sample from a patient. Then, for establishing a test level of ADCC exceeding a basic level, one or a plurality of lymphokines are given to the patient at least once and then the first aliquot of the effector cell taken out of the patient at a specific interval is measured at each interval. Then, for determining the change in NK cell activity that is higher than the basic level, a second aliquot of the effector cell is measured at each interval, thus establishing the level of the activated NK activity. Then, the reinforcement time within the optimum organism of effector cell is determined for the ADCC response.

摘要： PURPOSE: To prepare a composition capable of relatively increasing the amount of an active agent delivered to a target cell as compared with that delivered to a nontarget cell by attaching a diagnostic agent or a therapeutic agent to each of plural antibody species having non-overlapping patterns of cross- reactivity. CONSTITUTION: This functionally specific antibody is obtained by attaching the same or different diagnostic agents or therapeutic agents to each of antibody species in which each of two or more antibodies is reactive with different epitopes on a target site and patterns of cross-reactivity of each are non- overlapping. The antibody is capable of minimizing or eradicating the additive accumulation of an active agent in a nontarget tissue and additively accumulating the active agent in the target site. In the case of the diagnostic agent, the target site can be imaged against a relatively low background to reduce the possibility of misdiagnosis by using the composition. In the case of the therapeutic agent, the toxicity to normal tissues can be reduced.
