摘要： PROBLEM TO BE SOLVED: To obtain the subject new virus gene useful for ascertainment of the disease shape of G-type hepatitis, determination of administration course of therapeutic agent, etc., comprising a gene having a base sequence coding a protein of a specific amino acid sequence, shown by a base sequence of its cDNA. SOLUTION: This new G-type hepatitis virus gene has a base sequence which codes a protein of any amino acid sequence selected from amino acid sequence of formula I, formula II, etc., and is shown by a base sequence of its cDNA. The gene is capable of facilitating specification of the base sequence of a gene of a G-type hepatitis virus characteristic to Japanese, ascertainment of the disease shape of a G-type hepatitis, etc. The gene is obtained by separating an RNA from a virus particle from serum of patient of non-A, non-B and non-C type hepatitis, preparing a cDNA library by using the RNA, screening the library by using a probe having a partial base sequence of the gene and recovering a DNA from a plaque hybridized with the probe.

A region deciding means 34 decides in which region a blood cell belongs based on an A/D converted light intensity data corresponding to the number of CD55 and CD59 in one blood cell and means 35-37 accumulate the number of blood cells in each region. Subsequently, means 40-42 for deciding presence/absence of I-III region calculate the ratio of the total number of blood cell set in a measuring number setting means 39 to the total number of measured blood cells and output the ratio in a visible state.

The inlet part is heated to dissolve the fluorescent probe from the target polynucleotide and the probe crosses the a certain zone illuminated with a laser beam in the electrophoretic column to detect the fluorescence. The target polynucleotide of 100 copy order can be detected.

The deformability of the erythrocyte is determined based on the change of the differential pressure. A device for this purpose is provided with the filter 2 having a large number of hole portions of a diameter controlling the passage of the erythrocyte, a measuring tube 1 equipped with tube portions having the approximately same inner diameter with a filter diameter on the upstream side and the downstream side of the filter 2 respectively, the high sensitivity pressure meter 9 detecting the differential pressure between the upstream side and the downstream side, and a computing portion determine the erythrocyte deformability based on the determined differential pressure.

SOLUTION: The hybridomas are those originating from immunocytes of animals immunized with a gene containing a base sequence coding for an antigenic determinant whose gene is preferably modified in such a way that gene products take a form so as to be extracellularly exposed. These hydridomas are obtained, e.g. by immunizing BALB/c mice with a plasmid DNA (pRc/CMV-T48) integrated with the whole length of Th1 gene, and collecting them from splenic cells and myeloma cells of the immunized mice according to a well-known method. The objective monoclonal antibody can be easily obtained from these hybridomas by searching (e.g. immuno-blotting) for hybridomas that can produce a monoclonal antibody specific to the target gene products and culturing monoclonal cells on a large scale.

SOLUTION: This hepatitis C virus binding protein has the following properties. (1) It has a molecular weight of 2.5kDa (k-Dalton), (2) it can be bound to IRES of hepatitis C virus gene, (3) the translation efficiency of hepatitis C virus gene is remarkably depressed when the protein is not bound to IRES of hepatitis C virus gene, (4) it does not bind to the IRES of a mutant hepatitis C virus gene having a base sequence constituting the stem of α-branch structure (nt47-67) substituted with another base sequence and (5) it can be bound to the IRES of a mutant hepatitis C virus gene having a base sequence constituting the loop of α-branch structure (nt47-67) substituted with another base sequence.
