摘要： PROBLEM TO BE SOLVED: To provide a microwell array chip capable of making a time for preparatively isolating a cell short, without requiring a complicated device, capable of recognizing that an antigen binds to the cell, capable of detecting an antigen-specific lymphocyte of which the frequency is low (0.001% or more), and further capable of analyzing whether the lymphocyte to which the antigen binds reacts with another antigen or not. SOLUTION: This microwell array chip has a plurality of microwells, receives one of test lymphocyte in each of the microwells, and is used so that the antigen-specific lymphocyte is detected one by one as a unit. Further, the microwells each have such a shape and a size that only one of the lymphocyte is received by one of the microwells. Furthermore, the microwell array chip has a plurality of the micowells of which the each contains one of the test lymphocyte and is used in detecting the antigen-specific lymphocyte in such a manner that the antigen is added to each of the microwells to stimulate the cell and the cell which reacts with the antigen is detected. The method comprises the steps of: forming a film on at least one principal surface of a substrate; applying a resist coating on the film that has been formed; exposing the resist surface through a mask having a microwell pattern and removing uncured portions of resist; etching the exposed portions of said film and substrate to fabricate wells in the form of a microwell array; and removing the resist. A microwell array chip made of silicon and having multiple microwells, each microwell being used to store a single specimen organic cell, wherein each microwell is of a size and shape holding just one organic cell.

The antigen-specific immunoglobulin gene is cloned, when the antigen-specific lymphocyte comprises a B-lymphocyte, and the antigen-specific T-lymphocyte receptor gene is cloned, when the antigen-specific lymphocyte comprises a T-lymphocyte. The method for producing the monoclonal antibody comprises using the cloned antigen-specific immunoglobulin gene.
