摘要： Nucleic acid constructs and methods for rendering modifications to a genome are provided, wherein the modifications comprise null alleles, conditional alleles and null alleles comprising COINS. Multifunctional alleles (MFA) are provided, as well as methods for making them, which afford the ability in a single targeting to introduce an allele that can be used to generate a null allele, a conditional allele, or an allele that is a null allele and that further includes a COIN. MFAs comprise pairs of cognate recombinase recognition sites, an actuating sequence and/or a drug selection cassette, and a nucleotide sequence of interest, and a COIN, wherein upon action of a recombinase a conditional allele with a COIN is formed. In a further embodiment, action of a second recombinase forms an allele that contains only a COIN in sense orientation. In a further embodiment, action by a third recombinase forms an allele that contains only the actuating sequence in sense orientation.

摘要： Transgenic non-human animals one having a general deletor construct and a second having a general reporter construct are described. The general deletor animals express a heterologous recombinase under the control of a ubiquitously expressed endogenous promoter. Specifically, the Cre recombinase is inserted into the ROSA26 locus of the mouse. The general reporter animals have a gene which is desired to remove flanked by sites recognized by the heterologous recombinase. This flanked sequence is operatively associated with a marker gene such that when the gene sequence flanked by sites recognized by the heterologous recombinase is excised, the reporter gene is expressed. When the general deletor mouse is crossed with the general reporter mouse the heterologous recombinase is expressed in essentially all cells of the resultant descendants under the control of the ubiquitous promoter. Expression of the recombinase results in the excision of the desired gene in essentially all cells of the descendant animals. A conditional deletor construct and its use to derive a conditional deletor animal are also described.

摘要： A DNA sequence (-426 to +28 base pairs) cloned from the probasin (PB) gene promoter region confers androgen regulation in cell culture and prostate specific expression in transgenic non-human eukaryotic animals. Various PB promoter fragments impart preferential regulation by androgens compared to other steroid hormones on fused transgenes. Alteration of the DNA sequences and/or combinations permits production of an (1) assay for androgenic or anti-androgenic materials, (2) transgenic non-human eukaryotic animals models for prostatic disease, (3) cell culture models for prostatic disease, and (4) treatment of human benign prostatic hyperplasia and human prostate cancer by gene therapy. This invention permits assays on agonist and antagonist of the androgen receptor or pathways that result in androgen action, testing materials for carcinogenicity of the prostate, testing drugs and gene therapy, or protection potential of materials on prostatic cells against prostatic disease.
