    • The present invention addresses the problem of providing: an image processing device, an in-focus position specifying method, and an in-focus position specifying program capable of specifying an in-focus position from a fluorescence image and allowing an expression level of a biological material over the entirety of a tissue specimen to be quantitatively analyzed. The present invention is provided with: an input means for inputting a morphology image representing the morphology of cells in a tissue specimen in which a biological material is stained by fluorescent nanoparticles and fluorescence images which represent the same range as that of the morphology image, which represent the expression of the biological material, and in which the focal planes are changed at a predetermined interval in the height direction of the tissue specimen; a first extraction means for extracting cell areas from the morphology image or the fluorescence images; an in-focus plane specifying means for specifying, as an in-focus plane, the focal plane being most focused for each of the cell areas; a coordinate specifying means for specifying coordinates in the in-focus plane of the cell area; a storage means for storing the in-focus plane and the coordinates; a second extraction means for extracting a fluorescent bright point area from a fluorescence image in a focal plane corresponding to the in-focus plane; and a calculation means for calculating the number of fluorescent nanoparticles or a brightness value in the fluorescent bright spot area.
