摘要： The present invention provides a peptide fragment or a series of peptide fragments having a cell death-inhibitory activity, having the amino acid sequence consisting of 103 amino acid residues at the C-terminal of selenoprotein P, or having said amino acid sequence with one or several amino acid residues therein being deleted, substituted or added, or having a partial sequence of either of the above amino acid sequences, a medicament for treatment comprising said peptide fragment or a series of peptide fragments, an antibody to said peptide fragment or a series of peptide fragments, and a method for screening a cell death-inhibitory activity using said peptide fragment or a series of peptide fragments. The preferable peptide fragment or a series of peptide fragments of the present invention has the amino acid sequences shown in SEQ ID NO: 1 and/or SEQ ID NO: 2 or has a partial sequence thereof.

摘要： A process for preparing a peptide fragment having a cell death-inhibitory activity is provided. In order to obtain a selenoprotein P fragment having a cell death-inhibitory activity, full-length selenoprotein P was reacted with various serine proteinases and the resulting fragments were electrophoresed and assessed for a cell death-inhibitory activity. A process for preparing a selenoprotein P fragment was established by identifying an enzyme that produced bands corresponding to a molecular weight of not more than 3.5 x 10 4 in electrophoresis. A process for preparing a peptide fragment having a cell death-inhibitory activity according to the present invention may be used for amelioration, treatment and prevention of diseases caused by cell death and for more efficient production of useful living substances.

摘要： Disclosed are a mutant transposon vector and a method for introducing a foreign gene into a cell. A mutant transposon vector having a nucleic acid fragment inserted therein, the nucleic acid fragment having the properties (i) and (ii) or the properties (i) to (iii) below; a foreign gene-expressing cell which is prepared by introducing the mutant transposon vector into the chromosomal DNA of an established cell or a cell capable of being individuated and then additionally introducing a foreign gene-expressing cassette into a recombinant system (e.g., Cre-Lox system); a recombinant animal created by using the cell (in the case where the cell capable of being individuated is used); and a method for producing a foreign protein from the cell or the recombinant animal: (i) comprising a 5'-TIR sequence and a 3'-TIR sequence of transposon gene; (ii) having a sequence at which a recombination reaction occurs inserted between at least one set of DR regions of two sets of DR regions present in a 5'-TIR sequence or a 3'-TIR sequence of transposon gene; and (iii) having a restriction recognition site or a foreign gene-expressing cassette inserted between a 5'-TIR sequence and a 3'-TIR sequence of transposon gene.

摘要： A modified transposon vector and a method for introducing a foreign gene into a cell are provided. A modified transposon vector wherein a nucleic acid fragment having the following features (a)-(b) or (a)-(c) is inserted; a cell expressing a foreign gene obtained by introducing said vector into chromosomal DNA of a cell line or an ontogenetic cell and further introducing an expression cassette of a foreign gene into the chromosomal DNA; a genetically recombined animal generated by using said cell (in case of an ontogenetic cell); and a method for producing a foreign protein from said cell and said genetically recombined animal:
(a) It consists of 5'- and 3'-TIR sequences of a transposon gene; (b) A sequence where a recombination reaction occurs is inserted between at least either of DR regions present by twos each in 5'- and 3'-TIR sequences of a transposon gene; (c) A restriction enzyme recognition site or an expression cassette of a foreign gene is inserted between 5'- and 3'-TIR sequences of a transposon gene.

摘要： A novel selenocysteine-containing protein is provided which is a substantial entity having an enzymatic activity, typically an activity to reduce peroxide phospholipids. There are provided a gene consisting of a coding sequence for a protein not containing selenocysteine in which the codon for selenocysteine is introduced at a desired position and a selenocysteine insertion sequence resided at the 3'-end of said coding sequence and a protein expressed therefrom, typically a selenocysteine-containing protein having an activity to reduce peroxide phospholipids which is prepared by introducing selenocysteine into albumin and as well as a gene encoding said protein. A novel anti-oxidative substance is provided which can be applied for prevention of worsening, prophylaxis or treatment of various diseases associated with oxidative stress.
