    • PURPOSE: A cell proliferation promoting agent containing selenium and a method for preparing mesenchymal stem cells derived from a fetus in amniotic fluid are provided to lower the cost for preparing the mesenchymal stem cells and to obtain the mesenchymal stem cells with excellent effect on cell growth and skin regeneration ability. CONSTITUTION: A cell proliferation promoting agent contains selenium as an active ingredient. The cell is a mesenchymal stem cell. A method for preparing the mesenchymal stem cells derived from a fetus in amniotic fluid comprises a step of culturing amniotic fluid-derived cells in a medium containing 5-10% FBS and 1-20 ng/ml of selenium. The medium additionally contains bFGF(basic fibroblast growth factor). A method for preparing the medium for culturing the mesenchymal stem cells comprises: a step of sub-culturing the amniotic fluid-derived cells in the medium containing FBS and selenium and collecting the mesenchymal stem cells; and a step of culturing the mesenchymal stem cells in a serum-free medium for 1-10 days and preparing a conditioned medium.

    • PURPOSE: A method for preparing insulin-secreting cells through reprogramming from labium minor-derived adult cells is provided to easily and rapidly obtain massive adult cells from labium minor. CONSTITUTION: A method for inducing the differentiation of pluripotent adult stem cells which are derived from labium minor to insulin-secreting cells comprises: a step of culturing the adult stem cells in the media containing low glucose DMEM, 10% of FBS and 1% L-glutamine and 1% of penicillin-streptomycin; a step of culturing cultured adult stem cell in ITS media containing DMEM/F12(1:1), 5μg/ml of insulin, 50ug/ml of transferrine, 30nM of selenium and 1mM of glutamine; and a step of culturing the cultured cells in CM media to induce the differentiation. The CM media contains 10% of FBS (fetal bovine serum), 1% of L-glutamine (cambrex), 1% of penicillin-streptomycin, 0.5μM of hydrocortisone, 5 μg/ml of insulin and 10 mM of nicotinamide.
