摘要： A method is described for generating binding molecules. A defined population of binding molecules is produced using at least the following steps: (a) providing a population of B cells expressing a limited VL repertoire wherein essentially all B cells carry at least one VL; (b) obtaining nucleic acids from said B cells; (c) optionally, amplifying nucleic acids encoding immunoglobulin heavy chain variable regions in said sample; (d) at least partial sequencing of essentially all obtained nucleic acids of step (b) or amplification products of step (c); (e) performing frequency analysis of sequences from step (d); (f) selecting desired VH sequences; (g) providing a host cell with at least one vector comprising at least one of said desired VH sequences and/or at least one VL sequence of said limited VL repertoire; (h) culturing host cells and allowing for expression of VH or VL polypeptides; and (i) obtaining said binding molecules.
