摘要： PURPOSE:To obtain a new condensed pyrimidine derivative of glutamic acid bond having antitumor activity. CONSTITUTION:A compound of formula I [R is OH or NH2; R is phenylene, pyridinediyl, thienediyl, furandiyl or thiazolediyl; Co2R and CO2R are carboxyl which may contain protecting group; X-Y-Z is N-CH2-CH2, N-CH-CH or N-CH2- CH2; A is O, CR R (R and R are halogen, hydrocarbon which may be replaced or bonded to form alkylidene which may be substituted) or NR (R is H or hydrocarbon group); n is 1-3] or its pharmaceutically acceptable salt such as N-{4-[2-(2,4-diamino-6,7-dihydro-5H-cyclopenta[d]pyrimidin-5-yl)ethyl) benzoyl}-L- glutamic acid. The compound is obtained by reacting a compound of formula II or a reactive derivative of its carboxyl group with a compound of formula III.

摘要： PURPOSE:To obtain a new compound, having inhibitory action on a dihydrofolate reductase and useful as an antitumor agent CONSTITUTION:This compound is expressed by formula I [R is OH or amino; R is indolindiyl, indoldiyl, indandiyl or indendiyl; (n) is 0-4], e.g. ethyl 1-[2-(2,4- diamino-6,7-dihydro-5Hcyclpenta[d]pyrimidin-5-yl)ethyl]indole-5-carbox ylate. The compound expressed by formula I is obtained by condensing a compound expressed by formula II or a reactive derivative at the carboxyl group thereof with a compound expressed by formula III (R and R are carboxyl-protecting group), providing a carboxylic acid ester derivative expressed by formula IV and then hydrolyzing the resultant ester derivative in the presence of an acid or an alkali or removing the protecting groups of the carboxyl groups according to the hydrogenolysis including catalytic reduction.
