    • Total bilirubin or direct bilirubin is determined by reacting a biological fluid sample with nitrous acid as an oxidizing agent and measuring optical changes in the sample. To determine direct bilirubin, the sample is reacted with nitrous acid in the presence of thiourea which suppresses the reaction of indirect bilirubin. To determine total bilirubin, the sample is reacted with nitrous acid in the presence of cetyltrimethylammonium bromide which promotes the reaction. Alternatively, the biological fluid sample is reacted with an oxidizing agent in the presence of a specified nonionic surfactant to thereby determine direct bilirubin. These determination methods well correlate to the conventional enzymatic methods. Moreover, they are highly safe methods for determining bilirubin because of being excellent in the stabilities of reagents and causing little environmental pollution.
