    • PROBLEM TO BE SOLVED: To provide a method for isolation and purification of a highly anionic target protein such as a sulfated protein.SOLUTION: The method for purifying the highly anionic target protein comprises (1) a step to contact an ion-exchange chromatograph with a specimen and bond the target protein to a substrate, (2) a step to wash the substrate with a first washing solution under a condition to keep a plurality of proteinaceous and non-proteinaceous impurities in the specimen from bonding to the substrate or wash off the impurities from the substrate while leaving the highly anionic target protein in a state bonded to the substrate, (3) a step to elute the specimen with a first elution solution containing a salt solution having high molar concentration and (4) a step to collect the eluted specimen containing the purified anionic target protein. The sulfated protein has five or more sulfation sites, for example, six sulfation sites on N-terminal tyrosine residues such as PSGL-1 (P-selectin glycoprotein ligand).
