    • 1512228 Chromatography DURRUM INSTRUMENT CORP 23 Dec 1975 [25 June 1975] 52707/75 Heading B1H [Also in Division G1] In a continuous method for the detection of at least one primary amino acid and secondary amino acid accurring in a known sequence in a liquid sample stream moving in a flow path at a substantially constant flow rate to a detection zone, e.g. the element stream from a chromatographic column flowing to a fluormeter and wherein the detectability of the primary amino acid is impeded by reaction with an oxidising reagent while the dectability of the secondary amino acid is improved by reaction with said oxidising reagent, the steps of (a) adding an inert liquid diluent stream at a predetermined flow rate to the sample stream in the flow path when the sample stream includes a primary amino acid only at a selected location in the flow path, (b) adding stream containing said oxidising reagent at essentially by the same flow rate as the liquid diluent stream of step (a) at the selected location when the sample stream at the location includes a secondary amino acid only and simultaneously discontinuing the flow of the liquid diluent stream at that moment, whereby liquid flows through the detection zone at a substantially constant flow rate during detection of the primary and secondary amino acids, and (c) continuously adding a detection reagent to the sample stream flowing in the flow path downstream from the selected location, the said detection reagent being capable of quantitatively reacting with the primary and secondary amino acids in the stream to render the primary and secondary amino acids detectable. The detection reagent is preferably a fluorogenic reagent such as fluorescamine or o-phthalaldehyde, the oxidising agent may be N-chlorosuccinimide and the detection zone may be a fluorometer.
