摘要： A process for the primary clarification of feeds, including chemically treated flocculated feeds, containing the target biomolecules of interest such as mAbs, mammalian cell cultures, or bacterial cell cultures, using a primary clarification depth filtration device without the use of a primary clarification centrifugation step or a primary clarification tangential flow microfiltration step. The primary clarification depth filtration device contains a porous depth filter having graded porous layers of varying pore ratings. The primary clarification depth filtration device filters fluid feeds, including chemically treated flocculated feeds containing flocculated cellular debris and colloidal particulates having a particle size distribution of approximately about 0.5 pm to 200 pm, at a flow rate of about 10 litres/m2/hr to about 100 litres/m2/hr. Kits and methods of using and making the same are also provided.

摘要： The invention is to a method for purifying proteins using a bed of "cored beads" in a flow-through or polishing chromatography mode. Using a pellicular agarose/non-porous "cored bead" media provides one with media that have much improved permeability and mechanical properties over existing beads, while maintaining an increased protein "safety factor" and ease of scale-up over existing membrane devices. Cored agarose media use rigid internal beads (in this case beads of non-porous, synthetic polymer, metal or glass) which form incompressible and highly permeable packed beds especially when the agarose coating is relatively thin (e.g.5-15 microns on a greater than 50 micron diameter core). This enhanced permeability results in a lower protein capacity (especially in terms of g protein/ liter of media). Therefore, these materials can be used in applications where highly permeable beds are desired because one can tune the capacity, permeability and bed rigidity to maximize throughput or minimize processing time.

摘要： An exchange apparatus comprised of hollow thermoplastic tubes infusion bonded into a thermoplastic material is disclosed. In a preferred embodiment the hollow tubes are shaped by plaiting the tubes into cords and then thermally annealing the cords to set the crests and bends of the plait. 摘要： Hollow fiber membranes having a skinned surface on one diameter, and a porous surface on the opposite diameter are produced from perfluorinated thermoplastic polymers by extruding a heated solution of the polymer having a lower critical solution temperature directly into a cooling bath to form the porous membrane by liquid-liquid phase separation. Extrusion can be conducted either vertically or horizontally. The hollow fiber membranes are useful as ultrafiltration membranes and as membrane contactors.

摘要： A filtration cartridge having a housing with an inlet and an outlet and a filtration membrane contained within the housing and located between the inlet and the outlet is described. The membrane is sealed in such a manner that all fluid must flow through the membrane between entering the inlet and leaving the outlet of the housing. The entire cartridge, housing, membrane and any other components are all formed of one or more perfluorinated thermoplastic resins. These resins preferably are PFA, MFA, FEP and blends thereof. The membrane may be in the form of hollow fibers, wound fibers such as in a depth filter, or a flat sheet which may be formed into various configurations such as spiral filters, pleated filters, spiral pleated filters, disks and the like. These cartridges provide an inert, high temperature and acid/oxidative chemical resistant filter having properties similar to that of PTFE resins.
