摘要： The present invention relates to the identification of fibrinogen like protein 1 (FGL1) as a prognostic biomarker for responsiveness to an immune checkpoint inhibitor in a cancer patient. More specifically it relates to methods of treating cancer in a human patient comprising administering an immune checkpoint inhibitory antibody specifically inhibiting the interaction of fibrinogen-like protein 1 (FGL1) with lymphocyte-activation gene 3 protein (LAG3), wherein the cancer is an FGL1 expressing cancer. Further, the invention relates to methods for assessing susceptibility or predicting the responsiveness to the treatment with an immune checkpoint inhibitory antibody specifically inhibiting the interaction of FGL1 with LAG3 in a cancer patient, comprising detecting FGL1 expression in a sample from said patient and to a kit for selecting a cancer patient that would benefit from an immune checkpoint inhibitory antibody specifically inhibiting the interaction of FGL1 with LAG3. The immune checkpoint inhibitory antibody that specifically inhibits the interaction between FGL1 and LAG3 may be an antibody directed against human FGL1 or an antibody directed against human LAG3, optionally in combination with other immune checkpoint inhibitory antibodies, such as an anti-PD1 or an anti-PD-L1 antibody.

摘要： The present invention provides a method of treating melanoma and other cancers in a subject, the method comprising administering to the subject in need thereof a therapeutically effective amount of an herbal extract of PHY906 or Scutellaria baicalensis, a fraction thereof or any active chemical present in the herbal extract or fraction thereof, and one or more immune checkpoint inhibitors. In certain embodiments, the method further comprises administering at least one herbal extract, a fraction thereof or any active chemical present in the herbal extract or fraction thereof, of at least one herb selected from the group consisting of Glycyrrhiza uralensis, Paeonia lactiflora , and Ziziphus jujuba . The methods of the invention are useful for treating cancer such as melanoma, non-small cell lung cancer, renal cell carcinoma, liver cancer, colon cancer, urothelial bladder cancer and pancreatic cancer. In certain embodiments, administration of the herbal extracts of the invention counteract one or more gastrointestinal side effects of the immune checkpoint inhibitors and/or increase their therapeutic index against cancer.

摘要： The invention provides compositions and methods for regulating immune responses. In one embodiment, compositions of the invention include one or more SALM5-HVEM antagonists that are able to induce inflammation in the CNS or an immune privileged tissue. In another embodiment, compositions of the invention include one or more SALM5-HVEM agonists that are able to inhibit inflammation in the CNS or an immune privileged tissue. Therapeutic uses for SALM5-HVEM antagonists and agonist are also included in the invention.
