    • The invention describes a method of screening for factors that modulate gene expression, by detection of factors including factors that stimulate or inhibit cell growth, differentiation or proliferation, factors involved in one or more signal transduction pathways, or factors involved in protein-protein interactions. Identification of such factors that modulate gene expression is accomplished by detection of an intracellular event mediated by the factor. The intracellular event can be an increase or decrease in transcription or translational activities. Detection of the intracellular event is accomplished by any highly sensitive assay targeting a transcription or translation product, including, for example, a bDNA assay to the nascent transcript induced by the factor.

    • Novel polypeptides called signaling inositol polyphosphate 5-phosphatases are described called SIP-130, SIP-125, and SIP-N. SIP-130 is capable of binding to a PTB domain of SH2 and collagen containing protein (SHC). Also provided are polynucleotide sequences encoding the novel polypeptides, as well as vectors and host cells containing the polynucleotides. Further provided are modulators including agonists and antagonists of the novel polypeptides for use as therapeutics, including antibodies, polypeptides, small molecules, and polynucleotides and methods of using the therapeutics in treatment of diseases associated with abnormal cell growth. Methods of making the polypeptides, polynucleotides, vectors, host cells, antibodies, and small molecules are also provided. Gene delivery vehicles including SIP and SIP activity-modulating polynucleotides are described.

    • A nucleic acid molecule is provided that can be expressed in a host cell to produce a biologically active ob polypeptide that can effectively inhibit food intake and/or weight gain. Vectors and host cells containing the nucleic acid molecule are also provided, as well as methods for producing the ob protein and other ob polypeptides, methods of induction of the production of the ob polypeptides, such as by in vivo or ex vivo gene therapy, and methods for inhibition of food intake and/or weight gain. A method for identification, detection, or isolation of an ob receptor is provided as well as methods for production of antibodies to the ob receptor.
