摘要： The instrumentation and methods described herein are based on imaging and measuring single cell dose response by fluorescent ion imaging that records live cell responses to drug doses with no apparent damage to the cells or toxic side effects. Dose response curves and other pharmacological parameters can be computed by imaging and measuring oscillation changes for each dose and each cell. Large variability of cellular responses under the same experimental conditions indicates that pharmacological benchmarks are determined by statistical distribution of cells and cellular physiology. The instrumentation and methods described herein provide a means of measuring statistical variability of cell populations for improved screening and development methods for drugs.

摘要： Disclosed are transgenic cells trasfected with pituitary tumor transfer gene (PTTG)-containing expression vectors and uses therefor. The nucleic acids may be operatively linked to a vector, optionally provided with control and expression sequences and/or being carried by a host cell. In one embodiment, the host cell is a thyroid stimulating hormone (TSH)-sensitive cell transfected with an expression vector comprising a DNA segment encoding functional PTTG, wherein the cell overexpresses PTTG in response to TSH. Also disclosed is an in vitro cell model for the study of genetic regulation mediated by PTTG in a mammalian cell wherein PTTG expression can be modulated by exposing the cell to TSH or estrogen. In one embodiment, the cell model is used to study the effect of PTTG expression on NIS expression or to modulate NIS expression.

摘要： Methods for observing the activity of compounds, cells, and cell components during a cellular event are provided. Such live cell observation allows one to assess the function and impact of various compounds and other molecules upon the biochemical pathways responsible for the event. Embodiments of the present invention describe the live cell observation of compounds' effects on PTTG during mitosis; specifically, the ramifications that such effects may have on treating aneuploidy and/or cancer. Alternate embodiments of the present invention describe the combination of the live cell method with high throughput screening, to efficiently identify and examine compounds for a desirable effect on a cellular event, such as compounds that may be effective in the treatment of aneuploidy or cancer.
