摘要： Methods and systems for capture object-based assays, including for determining a measure of the concentration of an analyte molecule or particle in a fluid sample, are described. The methods and systems may relate to high sensitivity detection of analytes, sometimes using assay conditions and sample handling that result in the capture and detection of a high percentage of the analyte molecules or particles in a fluid sample using relatively few capture objects. Apparatuses and methods for immobilizing capture objects with respect to assay sites, in some instances with unexpectedly high efficiencies are also described. Some such apparatuses involve the use of force fields and fluid meniscus forces, alone or in combination, to facilitate or improve capture object immobilization. Also described are techniques for utilizing a relatively high percentage of capture objects in an assay sample, such as by using disclosed sample washing techniques, imaging systems, and analysis procedures that can reduce capture object loss.

DNA or RNA from the sample may be fragmented and exposed to a first type of binding ligand and a second type of binding ligand that comprise nucleic acid sequences complimentary at least a portion of a sequence contained in the target DNA or RNA. At least a portion of the fragmented DNA or RNA associates with at least one of the first type of binding ligand and/or the second type of binding ligand, wherein the first type of binding ligand and second type of binding ligand comprises nucleic acid sequences complimentary to a different portions of a sequence contained in the DNA or RNA. A portion of the sample exposed to the binding ligands is analyzed to determine the number of fragmented DNA or RNA sequences.

摘要： Described herein are systems and methods for the detection of and/or determination of a measure of the concentration of analyte molecules or particles in a fluid sample. In some cases, the systems and methods employ techniques to reduce or limit the negative effects associated with non-specific binding events. Certain methods of the present invention involve associating the analyte molecules at least a first type of binding ligand and at least a second type of binding ligand, and spatially segregating the analyte molecules into a plurality of locations on a surface. The presence of an analyte molecule at or in a location may be determined by determining the presence of both the first type of binding ligand and the second type of binding ligand.

摘要： Methods and systems for the detection of analyte molecules are generally described. In some embodiments, a method may comprise using a stabilizing agent prior to, during, and/or after one or more steps of a detection assay. The stabilizing agent may serve to maintain and/or increase the detectable signal indicative of the analyte during one or more assay steps and/or until the signal is measured. In such cases, the stabilizing agent may reduce and/or prevent one or more phenomena associated with signal decay, such as, e.g., dissociation between, aggregation of, and/or denaturation of analyte molecules and/or detection molecules. In some embodiments, the stabilizing agent can be used to improve the sensitivity of new and existing assays with little or no adverse effect on specificity. The methods and systems, described herein, may be used for a plethora of applications, including the detection or quantification of low levels of analyte molecules.
