摘要： The present invention relates to the use of compounds for the treatment of disorders involving a protein kinase, preferably a tyrosine kinase. 摘要： Described is a method for the identification of mutations in a receptor tyrosine kinase leading to a change in activity, said method comprises the steps of providing a host cell comprising a nucleic acid sequence derived from tissue, in particular cancerous tissue, of a particular individual said nucleic acid sequence encoding a polypeptide which comprises an active part of a receptor tyrosine kinase under the control of an inducible promoter, and determining the activity level of said RTK or rather the deviation of the activity from a standard form of the respective RTK, e.g. the wild-type (wt) RTK. Host cells prepared and characterized in the scope of the above defined method may then be used to determine the responsiveness to specific RTK inhibitors and/or to find new RTK inhibitors.

摘要： An in vivo method for the identification and/or validation of receptor tyrosine kinase inhibitors is described. Said method is characterised by the following steps: a) providing host cells comprising a nucleic acid construct encoding a peptide which comprises a tyrosine kinase domain of a receptor tyrosine kinase wherein said peptide lacks a transmembrane domain or a functional fragment thereof and said tyrosine kinase activity in the cytoplasma leads to proliferation arrest, b) contacting said host cells with a candidate compound and c) identification of inhibitors of said tyrosine kinase activity by cultivation of said host cells under suitable conditions such that the modulation of the tyrosine kinase activity by the candidate compound leads to cell growth.
