摘要： A process of preparing articles having gaseous NO sequestered therein is disclosed, as well as articles prepared thereby. Also disclosed are articles having gaseous NO sequestered therewithin and having reduced amount of oxygen-containing and/or nitrogen-containing reactive species. Also disclosed are processes of preparing packaged articles having a non-gas permeable package that comprises gaseous NO therein and packaged articles made therefrom. Also disclosed are charging devices which can be utilized in the above-described processes. The articles prepared by the above-described processes are preferably medical devices such as indwelling catheters, intubation devices and tampons. Tampons having sequestered therein gaseous NO, uses thereof and processes of preparing same are also disclosed.

摘要： Antimicrobial and biofilm-inhibiting nitric oxide-releasing medical appliances for installation into a patient, and processes for producing the medical appliances. The medical appliances are configured for conveying materials into and out of the patient's body and may comprise tubings or pouching systems. The medical appliances comprise a gas-permeable cured resin material selected from the group consisting of curable silicones, polyvinyl acetates, thermoplastic elastomers, acrylonitrile-butadiene-styrene copolymer rubber, polyurethanes and selected combinations thereof, wherein the gas-permeable cured resin material comprises a matrix suitable for releasably sequestering therein permeating gases. A plurality of nitric-oxide gas-releasing moieties is sequestered within and through the gas-permeable resin material. Nitric oxide is released from at least a portion of said plurality of nitric oxide gas-releasing moieties upon contact of the medical appliance with a moisture source. Exemplary medical appliances include endotracheal tubes, crichotracheotomy tubes, urinary catheters, wound drainage tubes, central intravenous catheters, peripheral intravenous catheters, and poucing systems.

A new method of developing drugs for treating M. tuberculosis is provided. Unique kinases (PknB, PknG, PknH, PknJ) to the organism are used to discover compounds capable of inhibiting them. Knockout mutants of the respective kinases display slower growth and viability both in vitro and in vivo. These observations are particularly noticeable in the case of pknG mutants.

摘要： The present invention utilizes three families of bacterial enzymes, which play a key role in mycothiol biosynthesis. The three families are bacterial cysteine:glucosaminyl inositol ligases (MshC) with catalytic ligase activity for ligation of glucosaminyl inositol and cysteine, bacterial acetyl-CoA:Cys-GlcN-Ins acetyltransferases (MshD) with catalytic activity for addition of an acetyl group to Cys-G1cN-Ins and bacterial MshA glycosyltransferase with catalytic activity for production of GlcNAc-Ins. The invention provides methods for using the mycothiol biosynthesis ligases, acetyltransferases or glycosyltransferases in drug screening assays to determine compounds that inhibit activity. The invention provides for treatment of actinomycete infections in mammals using antibiotics that inhibit production or activity of the enzymes of mycothiol biosynthesis, in particular MshC, MshD or MshA, and thereby reduce the production of mycothiol and the virulence of the infecting bacteria. Additionally, the invention provides a live mutant with a genome containing a modification in an endogenous enzyme of mycothiol biosynthesis gene. The invention also provides an expression vector comprising polynucleotides of mshA, mshB, mshC and mshD .
