摘要： An apparatus is provided for culturing and transfecting cells using a liquid. A bioreactor includes a cell culture bed for receiving the liquid. An external loop is connected to the bioreactor and adapted for circulating the liquid to and from the bioreactor. The external loop is adapted for introducing a gas into the liquid therein and/or for circulating a transfection material in the liquid therein to the bioreactor. In one embodiment, a first vessel is associated with the external loop for receiving the liquid, and a gas injector is provided for injecting a gas into the liquid, such as into the first vessel. A second vessel is also provided for providing a transfection material (such as a transfecting agent as part of a transfection mixture) to the liquid, such as directly to the bioreactor or to the external loop. In another embodiment, the gas injector is associated directly with the external loop. The external loop may be dedicated for gas injection and/or transfection, and maintained separate from a loop for circulating liquid media to the bioreactor, which potentially allows for operation on demand. Related methods are also disclosed.

摘要： The disclosure provides a system and a process for injecting and uniformly distributing a liquid into a fermentation broth in a bioreactor for maintaining stable condition within the fermentation broth. The system includes a downcomer section which contains two or more circular pipe conduits extending from a wall of the downcomer to the center vertical axis line of the downcomer. Liquid injection apertures are arranged along the circular pipe conduit and are non-uniformly spaced apart from the center vertical axis of the downcomer to the wall of the downcomer. The apertures further alternate and are offset between the top and bottom surfaces of the circular pipe conduit. The process of the disclosure involves using the system described above for carrying out a fermentation process for converting a C1-substrate to at least one product such as ethanol.

摘要： An inverted conical bioreactor is provided for growing cells or microorganisms. The bioreactor has an internal space and a perforated barrier within the vessel, through which a liquid may flow, where cells or microorganisms cannot pass through the perforated barrier. The perforated barrier divides the internal space of the bioreactor into a first chamber and a second chamber. Cells are grown within the second chamber and can be perfused by re-circulating the liquid, for example a growth medium, through the bioreactor. Various inlet ports and outlet ports allow controlling the parameters of flow of the growth medium.

The first vertical circulation loop is characterized by an individual loop mass transfer coefficient. A controller is coupled to the plurality of spargers and configured to control the plurality of spargers together such that a cumulative mass transfer coefficient of the plurality of vertical circulation loops is within a threshold of the individual loop mass transfer coefficient associated with the first vertical circulation loop.
