The method comprises(a) replacing the genomic copy of the target gene with a selectable marker gene through homologous recombination to form a first recombinant strain; and(b) replacing the selectable marker gene in the first recombinant strain with an altered copy of the target gene through homologous recombination to form a second recombinant strain.

    • The invention teaches means for detecting unauthorized tampering or substitutions of a device, and has particular utility when applied on a "seal" device used to secure a location or thing. The seal has a transparent body wall, and a first indicia, viz., a label identification is formed on the inside surface of this wall. Second and third indicia are formed on the outside surface of the transparent wall, and each of these indicia is transparent to allow the parallax angled viewing of the first indicia through these indicia. The second indicia is in the form of a broadly uniform pattern, viz, many small spaced dots; while the third indicia is in the form of easily memorized objects, such as human faces, made on a substrate by means of halftone printing. The substrate is lapped over the outside surface of the transparent wall. A thin cocoon of a transparent material, generally of the same material as the substrate such as plastic, is formed over the seal body and specifically over the transparent wall and the second and third indicia formed thereon. This cocoon is seamless and has walls of nonuniform thickness. Both the genuineness of the seal and whether anyone has attempted to compromise the seal can thus be visually determined upon inspection.

    • A steam trap monitor positioned downstream of a steam trap in a closed steam system includes a first sensor (the combination of a hot finger and thermocouple well) for measuring the energy of condensate and a second sensor (a cold finger) for measuring the total energy of condensate and steam in the line. The hot finger includes one or more thermocouples for detecting condensate level and energy, while the cold finger contains a liquid with a lower boiling temperature than that of water. Vapor pressure from the liquid is used to do work such as displacing a piston or bellows in providing an indication of total energy (steam+condensate) of the system. Processing means coupled to and responsive to outputs from the thermocouple well hot and cold fingers subtracts the condensate energy as measured by the hot finger and thermocouple well from the total energy as measured by the cold finger to provide an indication of the presence of steam downstream from the trap indicating that the steam trap is malfunctioning.

    • The invention is a tool to aid in the attaching of coaxial electrical cable ends to a half of a coaxial cable connector assembly. In detail, the tool comprises a wrench to prevent rotation between the connector halves when the connector halves are joined. A shaft is included having a threaded end for coupling to the recess of the other connector half. Finally, a drive preferably in the form of a portable electrical drill, is used to rotate the drive. Thus, with the joined connector halves installed in the wrench, so that rotation therebetween is prevented, the shaft can be threaded into the other connector half and thereafter coupled to the drill. The connector half to which the coaxial cable is to be joined is aligned with the prepared cable end and fed into the threaded recess as the wrench and joined connector halves are rotated.

    • A system and method for pre-charging the DC bus of a utility connected power converter. In one embodiment, a full-wave rectifier circuit is coupled to the utility grid through an isolation transformer and circuit protector. The rectifier circuit output is connected to the DC bus of the power converter to provide a pre-charging current to the DC bus capacitors. The output from the rectifier circuit also supplies power to the control circuit power supply for the converter. When the control circuit closes the contactor of the converter system and activates the converter, the voltage across the DC bus capacitors is increased above the level of the rectified power grid voltage provided at the output of the pre-charge circuit. This reverse-biases the rectifier diodes, placing the pre-charge circuit into an inactive state. The impedance of the isolation transformer limits the in-rush current into the DC bus capacitors during the pre-charge.
