    • The present invention is directed to a fluid-imbibing dispensing device for the immediate or almost immediate and extended delivery of an active agent over a prolonged period of time together with the initially delayed pulse delivery of an active agent to a fluid environment of use. The dispenser comprises a housing having a first wall section and a second wall section in slidably telescoping arrangement with each other, which housing maintains its integrity in the environment of use; an internal compartment surrounded and defined by the housing; a first active agent delivery chamber within a portion of the compartment defined by the first wall section, the first delivery chamber including at least one active agent formulation, an exit means for providing communication between the active agent formulation and the environment, and a first expansion means for dispensing the first active agent formulation through the exit means to the environment immediately and over a prolonged period of time; a second active agent delivery chamber within a portion of the compartment defined by the first wall section and adjacent the open end of the first wall section, the second delivery chamber including at least one active agent formulation; and an expansion chamber within a portion of the compartment defined by the second wall section, for separating apart the first and second wall sections of the housing after exposure to the environment of use for dispensing the second active agent formulation to the environment in a pulse after an initially delayed period of time, the expansion chamber including a second expansion means and a partition layer or push plate.

    • A diffusional drug delivery device is described which can provide for delayed onset of therapeutic effect and for delivery of the therapeutic agent in predetermined temporal patterns at optimum rates. Delay means are provided between the agent reservoir and the surface through which the agent is released by diffusion to delay the release of agent at a therapeutic rate for predetermined times after application. Suitable means include a delay membrane disposed between the releasing surface and the agent reservoir, which membrane is preferably free of undissolved agent and/or is initially impermeable to the agent and thereafter becomes permeable. One or more agent chambers defined by one or more of such membranes may be provided, whereby agents are released in predetermined temporal patterns at optimal release rates. Delayed permeability enables programmed washout periods to be obtained from the sequential and concurrent application of devices for the administration of drugs, such as nitrates, to which patients may develop a tolerance on continuous administration over extended time periods.

    • A drug formulation chamber for an intravenous administration set is provided. The intravenous administration set includes a container of an IV fluid, a drip chamber, a drug formulation chamber, and an adapter-needle assembly. The drug formulation chamber has a fluid inlet and a fluid outlet for maintaining a flow of IV fluid through the chamber. A portion of the chamber wall is comprised of a window material which allows the drug to diffuse therethrough but which prevents convective loss of the IV fluid. A flow distributor is provided within the chamber for distributing the flow of IV fluid along the interior surface of the window. A transdermal-type drug delivery device is adhered to the exterior surface of the window. Drug is delivered by the delivery device through the window and into the flowing IV fluid. The device delivers drug into the IV fluid at a rate that is independent of the flow rate of IV fluid through the formulation chamber. The rate of drug delivery from the device into the IV fluid is controlled by either the window itself or by a membrane layer in the drug delivery device. A plurality of drug delivery devices may be adhered to the window to deliver a plurality of drugs or to deliver a single drug at a higher dosage rate. A similar window may be placed in an IV bag to deliver a drug into the IV fluid therein.

    • A drug formulation chamber for an intravenous administration set is provided. The intravenous administration set includes a container of an IV fluid, a drip chamber, a drug formulation chamber, and an adapter-needle assembly. The drug formulation chamber houses a diffusional or an osmotically driven drug delivery device. The osmotically driven delivery device has a semipermeable, microporous or diffusional wall which surrounds a compartment containing the drug. Drug is delivered through an orifice in the semipermeable/microporous wall or through the diffusional wall itself, into the IV fluid. The device delivers drug into the IV fluid at a rate that is independent of the flow rate of IV fluid through the formulation chamber. The rate of drug delivery is controlled by variably adjusting the surface area of the semipermeable/microporous/diffusional wall that is exposed to the IV fluid flowing through the drug formulation chamber.
