摘要： A method of freezing a standard donor unit of red blood cells such that it can be stored for long periods and subsequently recovered in a form pure enough for transfusion purposes involves centrifuging a blood unit to remove plasma and platelets and to provide a Packed Cell Volume of the red blood cells of not less than 90%, adding the red blood cells to a freezing bag, containing HES solution such that the ratio of HES/red blood cell freezing unit is not more than 7% (preferably 6%) w/v, positioning the freezing bag in a freezing frame adapted to maintain the thickness of the contents of the bag constant, and placing the frame without shaking, into liquid nitrogen.

摘要： A cryogenic storage apparatus comprises a housing with an automatically operated access door. A holder device disposed in the housing supports a plurality of specimen-containing ampules, while a conveyor moves the holder device, together with the plurality of ampules, through cooling fluid in the housing along a path preferably including a snaking portion with vertically extending folds. An inverted L-shaped cooling component with an opened upper side is disposed in the housing for maintaining the cooling fluid at a predetermined low temperature. An extraction mechanism disposed outside of the housing at the access door serves to remove a selectable ampule positioned in the housing in juxtaposition to the door. A tracking device automatically tracks the positions of the ampules during motion thereof along the snaking path, while a control unit connected to the tracking device, the drive mechanism, the conveyor and the extraction mechanism activates the conveyor to move a given ampule along the path to the access door, opens the access door, and operates the extraction mechanism to remove the given ampule from the holder device and out through the opened door.

摘要： Disclosed are compositions and methods involving the use of PRG4 protein, also known as lubricin, to mechanically inhibit biological processes involving cell motility and adhesion. The methods and compositions may be used to develop a variety of specific therapies and compositions, often exploited through surgical procedures, where development of the pathology involves one or more of the following modes of action: 1) the passage of cells from one body compartment to another, 2) adherence of macrophages to substrates such as fibrin or exposed extra cellular matrix, 3) binding of platelets to fibrin, or 4) failure of function of the glycocalyx on exposed epithelial cell surfaces, e.g., within the vasculature. In these instances PRG4 glycoprotein adheres to extracellular matrix or cell surfaces and presents a glycol-surface of polysaccharide which blocks the mechanisms of cell motility, extravasation, or intravasation, inhibits sticking of macrophages and platelets, and/or serves as a substitute or mimic of native glycocalyx.
