CONSTITUTION: Pseudomonas sp.CU1(KCTC 18065P) is characteristically isolated by the steps of: inoculating the soil contaminated with crude oil into a minimal medium added with oil as a carbon source then culturing the medium at 20-30 deg.C; smearing the cultured medium on nutrient agar medium to isolate microorganisms for degradation of crude oil; and selecting a strain showing excellent growth and oil degradation.

The microorganism is useful for the bioremediation of lubricant contaminated soil and underground water. CONSTITUTION: The novel microorganism, Bacillus sp. LU1 strain(KCTC 18064P) degrades a lubricant which is non-degradable hydrocarbon consisting of C18-15, at 10-30 deg.C, pH 5-9, preferably at 20-25 deg.C, pH 6-7. It is separated by the steps of: culturing oil contaminated soil in a minimal medium containing a lubricant; smearing the cultured medium on a nutrient agar medium; and selecting a microorganism having excellent lubricant degradability.

CONSTITUTION: The psychrophile microorganism Pseudomonas sp. CX3B(KCTC 18054P), which is capable of decomposing oil including diesel below 12 deg.C., is isolated from the soil. Its preparation process comprises the steps of: collecting samples from the various oil-contaminated soils; inserting the samples into a phosphate buffer solution, agitating and sonicating them at 4 deg.C for 1 minute; cultivating the sample solution in a medium containing diesel and crude oil as a sole carbon source at 5 deg.C and 12 deg.C; spreading the cultivated medium on an agar medium to select oil-decomposing colonies through the color change by 2,6-DCPIP(Dichlorophenol-Indophenol) reduction; and screening a microorganism showing the most excellent oil-decomposability and viability at low temperature.

    • PURPOSE: Provided is a strain Pseudomonas sp. D3K1 (KCTC 8935P) isolated and purified from the soil contaminated by oils. Also, a method for preparing the strain. The strain is useful for recovering the soil and purifying the water because it is excellent in decomposing the oils and carbohydrates. CONSTITUTION: The strain Pseudomonas sp. D3K1 is isolated and purified by the steps of : (1) obtaining the soil contaminated by oils; (2) isolating the strain decomposing oils, wherein the sample is pre-treated by mixing sterilized PBS and the soil in a ratio of 2:1 followed by homogenizing, ultrasonic desorbing and the sample is used as an inoculum in the culture; (3) screening the strain growing rapidly using oils as a unique carbon source by 2, 6-DCPIP (2, 6-dichlorophenolindophenol) test; (4) identifying the screened strain using MIDI method.

C for 15 minutes at intervals of 1 minute; inoculating each supernatant into a minimal medium containing diesel and culturing them at 4 deg. C for 4 weeks; subculturing the samples; plating them on nutrient broth agar and culturing them to isolate colonies grown; inoculating the colonies into a minimal medium containing DCPIP and culturing them at 25 deg. C for 1 week; determining the color change of the cultivated medium; and selecting microorganisms showing excellent growth activity.
