摘要： A composition comprising non-methylated CpG islands of a PGP9.5 gene promoter is provided to be used to diagnose cancer since the methylation of the PGP9.5 gene influences on the expression of the PGP9.5 in cancer cells. A composition for diagnosing cancer such as gallbladder carcinoma, non-small cell lung cancer, invasive colorectal cancer and esophageal squamous cell carcinoma includes a polynucleotide sequence consisting of a sequence of SEQ ID : NO. 1, wherein bases from 131st to 329th of the SEQ ID : NO. 1 consist of non-methylated bases. A genetic marker for diagnosing the cancer consists of 198-496 numbers of a consecutive base sequence including the bases from 131st to 329th of the SEQ ID : NO. 1. A method for detecting methylation of a PGP9.5 gene promoter comprises the steps of: (a) obtaining a sample from an individual; (b) obtaining a genome DNA from the sample; (c) treating the genome DNA with a compound capable of deforming a cytosine base or a methylation sensitive restriction enzyme; (d) amplifying the treated DNA using a primer pair consisting of SEQ ID : NOs. 4 and 5 or 6 and 7 capable of amplifying CpG islands of the PGP9.5 gene promoter; and (e) identifying the existence of an amplified product. A kit for diagnosing the cancer comprises a primer for MSP(methylation specific PCR) consisting of a primer pair of SEQ ID : NOs. 4 and 5 as an effective ingredient.
