A biosensor for detecting a material in a biological sample electrochemically comprises: an insulating lower substrate(100) containing at least one electrode(111, 112, 113), and an enzyme reaction layer(120) for reacting with the material in the sample on the surface; a spacer(600) for attaching the lower substrate to the upper substrate, and forming a sample introduction space(610) for inducing absorption of the sample to the position of electrode along the enzyme reaction layer; and an insulating upper layer(700) having an air release space(710) for releasing the air absorbed together with the sample through the sample introduction space, wherein the air release space has a three-dimensional shape formed in a different layer from the sample introduction space. Further, a section of the air release space has 0.3 to 3 mm of width and 0.1 to 3 mm of height.
