摘要： PROBLEM TO BE SOLVED: To simply determine the L-carnitine content as an indicator for ascorbic defection with high reproducibility by admixing a specific substance to a carnitine acyltransferase reaction system. SOLUTION: A sample solution containing L-carnitine is incubated with carnitine acryltransferase in the presence of 2,6-dichlorophenol-indopenol sodium salt (DCIP) and Co A in a buffer solution of 6-9 pH to measure the maximum decrease in the visible extinction. L-Carnitine is determined by comparing the determined value with the calibration curve prepared by measuring known L- or D-carnitine under the same conditions.

摘要： PURPOSE:To provide a senescence-preventing agent containing an RV substance produced from micro organism belonging to the genus Rhizopus as the active component, showing a hypophysis-stimulating effect via the diencephaloc system, capable of preventing accumulation of peroxides and effective for preventing senescence and cerebral apoplexy. CONSTITUTION:Rhizopus V-1 strain (Brkoken stipulation No.3646) belonging to the genus Rhizopus is aerobically cultured in a culture medium at 26-28 deg.C and pH 4-7 for 3-10 days and the resultant cultured solution is diluted with water, exudated with hot water and filtered. The filtrate is extracted with ether to remove ether-soluble components and the remainder ether-insoluble components are concentrated, thus obtaining the objective senescence-preventing agent having a composition of 21.5wt.% amino-state nitrogen compound, 33.0wt.% sugar and 3.4wt.% ash in a condition of 41.8wt.% water content, showing UV-absorption maxima at 210nm and 280nm, positive to ninhydrin reaction and buret reaction, stable at pH 2-12, resistive to treatment at 100-120 deg.C for 30min, soluble in water, slightly soluble in alcohol, colored white-pale yellow, having a hypophysis-stimulating actively and containing RV-substance derived from bacterium of the genus Rhizopus genus bacterium as the active component.

SOLUTION: The active component of the objective physiologically active substance expressing mitogen activity is a hot-water extract fraction obtained by fermenting a bran-barley bed with a microorganism of the genus Rhizopus, defatting the fermentation product with methyl alcohol, extracting the residue with water at room temperature and extracting the water-extraction residue with hot water of 100 deg.C. The hot-water extract fraction is separable by ion exchange chromatography into the 1st fraction detectable by phenol-sulfuric acid method and carbazole-sulfuric acid method and composed of a neutral sugar (detectable by phenol-sulfuric acid method) accounting for >=90% of the fraction and an acid sugar (detectable by carbazole-sulfuric acid method) and the 2nd fraction composed exclusively of an acid sugar detectable only by carbazole-sulfuric acid method.
