SOLUTION: This method for producing guanosines or 2-deoxyguanosines is carried out by acting purine nucleoside phospholylase on either one of ribose-1-phosphate or 2-deoxyribose-1-phosphate and glyoxal-guanine to form Glyoxal-guanosines or by acting purine nucleoside phospholylase and pyrimidine nucleoside phospholylase on either one selected from the group consisting of uridine, 2-deoxyuridine and thymidine and glyoxal-guanine in the presence of phosphoric acid ion to form glyoxal-guanosines and then carrying out the alkali decomposition of the glyoxal-guanosines.

摘要： PURPOSE:To obtain trifluorothymidine in high yield by base exchange reaction under a mild temperature condition of 30-45 deg.C using bacteria classified as Serratia sp. between trifluoromethyl uracil and deoxyribose donor. CONSTITUTION:Bacterial classified as Serratia sp. such as Serratia marcescens IFO 3052 are put to culture in a conventional medium containing a carbon source such as glucose and a nitrogen source such as ammonium sulfate, and the cultured bacteria are separated from the resultant culture solution. Thence, a reaction is made at 30-45 deg.C in an aqueous solution containing the above bacteria and phosphoric acid between trifluoromethyl uracil and a deoxyribose donor selected from 2'-deoxyadenosine and 2'-deoxyinosine. This reaction may be made using an immobilized enzyme. After completing the reaction, the bacteria or enzyme is separated from the system, and the trifluorothymidine produced is isolated and purified. Thus, the objective trifluorothymidine can be obtained continuously in high yield.

CONSTITUTION:An aqueous solution containing (1) a ribonucleoside selected from inosine, adenosine, guanosine and uridine or a deoxyribonucleoside selected from 2'-deoxyinosine, 2'-deoxyadenosine, 2'-deoxyguanosine and 2'-deoxyuridine, (2) thymine and (3) phosphoric acid or phosphate, is put to base interchange reaction in the presence of Bacillus thermophile or treated product therefrom, thus obtaining the objective nucleoside compound as 5-methyl uridine or thymidine. The base interchange reaction temperature is pref. 40-70 deg.C.

Therein, the produced pyrimidine base is converted into a compound with an enzyme originated from a microorganism. The compound can not be converted into the substrates of pyrimidine nucleoside phosphorylase and purine nucleoside phosphorylase.

摘要： PROBLEM TO BE SOLVED: To obtain a xanthine dehydrogenase enzyme that has excellent thermal resistance and is useful for production of nucleoside compounds, etc., by a nucleic-acid base exchange reaction by culturing a microorganism belonging to the Pseudomonas genus having ability to produce the xanthine dehydrogenase. SOLUTION: This xanthine dehydrogenase enzyme is obtained by culturing Pseudomonas espy-Y-510 (FERM P-15104), a microorganism that belongs to the Pseudomonas genus able to produce xanthine dehydrogenase, which acts on hypoxanthine and xanthine so as to catalyze the reactions shown by the formulas in the pH range of 7-9, the most preferably around pH8.9, at an appropriate reaction temperature of 40-70 deg.C, the most preferably around 60 deg.C, without losing activity even when heated at 69 deg.C for 30min, and has a half life of the enzymatic activity of 25hr at 60 deg.C, a molecular weight of 14,1000 when determined by the gel filtration method, a Michaelis constant (Km) of 0.327mM to xanthine and 0.060mM to DNA, and an isoelectric point of 6.0 when determined by the isoelectric-point electrophoresis (pI).
