摘要： PURPOSE:To obtain a remedy for liver disorder useful for treating liver diseases such as acute and chronic hepatitis, etc., containing lignan as an active ingredient. CONSTITUTION:The titled remedy containing lignan shown by formula I as an active ingredient. The compound is obtained by extracting Schizandrae fructus with a warm lower hydrocarbon (e.g. petroleum ether or N-hexane), distilling away the solvent of the extracted solution to give residue, dissolving the residue in water, subjecting to steam distillation, subjecting unpurified oil part from which a purified oil is removed to chromatography using silica gel, isolating a compound shown by formula II by developing with n-hexane, benzene, acetone or a mixed solvent thereof, reacting the compound shown by formula II in benzene by the use of lead tetracetate, subjecting to the reaction product to preparative thin-layer chromatography after the reaction and purifying. A dose of the compound is 5-80mg per adult daily by oral administration and 0.1-10mg daily by parentral administration and properly administered dividedly several time.

PREPARATION:For example, HAKUGOMISHI (dried fruit of Schizandra chinensis Baill.) is extracted with lower hydrocarbons while warm and the solvents are then distilled away to provide a residue, which is subsequently dissolved in water, subjected to steam distillation and column chromatography using silica gel and developed with a solvent, such as n-hexane, to afford a compound expressed by formula II. The resultant compound expressed by formu la II is used as a raw material, incubated by adding a hepatic homogenate, reacted, separated and purified while confirming the compound expressed by formula I by thin layer chromatography to provide the aimed compound expressed by formula I.

The substance can be prepared into an oral drug such as tablet, capsule, granule, fine granule or powder or parenteral drug such as injection or suppository. A dose is 20-600mg calculated as weight of gomisin N per adult daily and administered dividedly several times in the case of oral administration and properly 1-60mg calculates as weight of gomisin N per adult daily in the case of parenteral administration.
