SOLUTION: This method for extracting and purifying a plant DNA comprises the following processes (a) to (e); (a) mixing a plant material with a solution containing a surfactant and, if necessary, a protein-denaturating agent to lyse cells in the plant material; (b) mixing the obtained lysed cell liquid with an organic solvent for extracting the surfactant to separate the aqueous phase from the organic solvent phase; (c) mixing the obtained aqueous phase with an adsorption liquid containing a chaotropic substance and a nucleic acid-binding solid phase carrier under a neutral to weakly alkaline condition to allow the nucleic acid-binding solid phase carrier to adsorb the plant DNA contained in the plant material; (d) washing the nucleic acid-binding solid phase carrier adsorbing the plant DNA with a washing liquid to remove saccharides and proteins contained in the plant cells; and (e) eluting the DNA bond to the nucleic acid-binding solid phase carrier.

The solid-phase carrier adsorbing the plasmid DNA thereon is then washed with a wash liquid and the plasmid DNA is eluted from the washed solid-phase carrier with an eluent to thereby extract and purify the plasmid DNA from the microorganism or cell in high purity for a short time without requiring complicated operations.

(5) Treating the membrane at temperature of 50-70 deg.C in a reaction liquid in which the labeled dioxynucleoside triphosphate is contained and DNA polymerase functions; (6) Removing excess dioxynicleoside triphoshate which remains in the liquid without being subjected to the reaction; (7) Detecting the labeled dioxynucleoside which has been taken in the membrane surface; and (8) Identifying the clone containing the gene on the basis of the detected signals.
