    • PURPOSE:To improve heat stability by culturing Pseudomonas sp. FH 1227 strain (Bikokenkinki no. 11618) (PMS) in a culture medium. CONSTITUTION:PMS strain is inoculated in a culture medium obtained by blending a carbon source such as glucose with a nitrogen source such as ammonium sulfate and inorganic salts such as NaCl and then cultured at 25-37 deg.C and pH 5.5-7.5 for 18-24 hr using the aeration agitation method, etc., to obtain a cultured material. The resultant cultured material is then subjected to centrifugal separation, etc., and the separated cells are ground using the ultrasonic grinding method, etc., to obtain a crude enzyme solution. The resultant crude enzyme solution is subsequently purified by ion-exchange chromatography, etc., thus producing the objective glucose dehydrogenase having following physicochemical properties; Action: catalyzing reaction of the formula; Substrate specificity: specific action on beta-D-glucose and 2deoxyglucose; Optimum pH: 8.5:9.0; Optimum temperature: 55 deg.C; pH stability: 6.0-7.5; Heat stability: stable to 50 deg.C at pH 7.0; Molecular weight: about 101000 determined by gel filtration method using TSK gel G3000SW; Isoelectric point: about pH 4.5; etc.
