摘要： PURPOSE:To obtain immunological agglutination reagent having high detection sensitivity and little non-specific agglutination by obtaining a sensitized carrier by sensitizing an antigen or an antibody to a carrier consisting of non-biological particles and then treating the sensitized carrier in non-polar amino acid solution. CONSTITUTION:A method for fabricating immunologically agglutinated particles comprises steps of obtaining a sensitized carrier by sensitizing an antigen or an antibody to a carrier consisting of non-biological particles and then treating the sensitized carrier in non-polar amino acid solution. To show carriers consisting of non-biological particles as examples, polystyrene latex, kaolin, carbon powder, organic-inorganic compound particle gelatin particle may be named. An antigen is a substance for producing an antibody to induce body fluid immunology or cell immunology, which may be for example protein, glycoprotein, lipid protein, lipid, nucleic acid, etc., wherein an antigen richer in hydrophobic property has a large effect in maintaining sensitivity, reduction in non-specific reaction, prevention of damage of an agglutinated image, etc. An antibody has activity for specifically coupling with an antigen, wherein IgG, IgM, IgA, IgD, IgE may be named.

摘要： PURPOSE:To produce a polypeptide having HCV antigenic activity by culturing Eschericha coli transformed with a recombinant vector prepared by integrating a structural protein gene of hepatitis C virus in a culture medium. CONSTITUTION:Escherichia coli is transformed with a recombinant vector prepared by integrating a structural protein gene of hepatitis C virus expressed by formula I. The resultant transformed Escherichia coli is then cultured in a culture medium to produce a polypeptide having HCV antigenic activity. The produced polypeptide is subsequently collected from the resultant culture. The structural protein gene of the hepatitis C virus can be inserted into the downstream side of a promoter of a vector utilizing a restriction enzyme site derived from a linker added in preparing the cDNA library, a restriction enzyme site derived from a plasmid in which the structural protein gene is inserted or a restriction enzyme site in the interior of the structural protein gene, etc. The obtained recombinant vector is capable of expressing the structural protein gene of the hepatitis C virus in the bacterial cell of Escherichia coli.
