摘要： PURPOSE:To provide an experimental instrument and an experimental device by which the behavior of tear liquid and aqueous humor during lotion dropping in the eyes is simulated in an almost actual state so as to measure the behavior of drugs accurately. CONSTITUTION:Experimental instruments 1a and 1b are desined as to simulate the state of cornea front part tear liquid layer or cornea rear part (aqueous humor) by pooling various kinds of solutions. The instruments are provided with a cell body 2, in which a stirring part 22 is formed on its bottom and whose outer circumference is covered with a water jacket 25, a sample membrane setting part 21 projecting on the outer circumferential face of the body 2 to set a sample membrane 5, a discharging tube 23 projecting on the circumferential face of the part 22 to discharge a solution in the body 2, an inlet tube 24 projecting on the upper side of the body 2 to flow a solution into the body 2, a magnet rotator 3 prepared within the part 22, and a magnet stirrer 4 prepared outside the body 2 to rotate a magnet rotator 3 within the body 2. In addition, an experimental device 10 is joined with two instruments 1a and 1b on the part 21 by means of the membrane 5.

摘要： PURPOSE: To predict and evaluate a penetration rate of a percutaneously administered drug through the stratum corneum according to a simple test in a short time by applying the drug to the surface of skin and then measuring the amount of the drug entering the stratum corneum under transient conditions twice at a time interval. CONSTITUTION: A drug (e.g. the one to be compounded in a percutaneous drug transport system such as a cataplasma, a plaster, a pasta or an ointment, applied to the skin surface and introduced through the skin into the body) is applied to the skin surface of a living body and the amount of the drug entering the stratum corneum is then measured twice under transient conditions at a time interval. The first measurement is preferably carried out after the passage of 10min from the application and the second measurement is conducted after the passage of 30min thereafter. The diffusion coefficient and the partition coefficient are determined on the basis of the measured amount of the drug in the stratum corneum to thereby determine the penetration rate of the drug in a steady state. The percutaneous penetration rate of the drug is then evaluated and predicted. The total amount of the drug entering the stratum corneum is determined by a stripping method using a tacky tape.
