Then, a sponge 6 is provided on a lower substrate 2 and impregnated with an anti-human-hemoglobin monoclonal antibody having a gold-colloid-particle label. The human hemoglobin is dropped on the sponge. Then, a connecting part 3 is bent, and the substrates 1 and 2 undergo contact bonding. The device is left alone for 5 minutes at room temperature. Then, both substrates are separated. The first and second regions are compared and observed.

By this method, even in a hue changing method accompanied by agglutination reaction and sandwich immunoassay, excellent sensitivity, stability and reactivity are obtained. When a particle size is set to 7 nm or less and 85% or more of colloid particles is adjusted to a uniform particle size, a further effective result is obtained.

Among these method, the glutaraldehyde treatment is more preferable in terms of ease and in terms of the efficient formation of self- polymn. The unreacted glutaraldehyde is neutralized by adding monoethanol amine thereto and is removed by dialysis with a large volume of 0.5% NaCl water as the simpler method.

The 1cm-square filter paper is stuck to the back side of the antibody adsorbing film, and it is put on a glass plate. 0, 0.1, 0.5, 1, 10mug/ml-human hemoglobin solutions 0.03ml are added and mixed to the anti-human hemoglobin monoclonal antibody-sensing gold colloid liquid 0.3ml set with the absorbance at 540nm to 50.D, 0.2ml of it is dripped on the antibody adsorbing nitrocellulose film and reacted for 5min, then the coloring of purple gold colloid grains is observed. It is judged as positive (-) if the coloring of the gold colloid grains is noticed, and it is judged as negative (-) if no coloring is noticed.

摘要： PURPOSE:To collect a large amount of a novel alkali phosphatase exhibiting an electrophoresis pattern shown by the figure and to use the phosphatase for diagnosing various kinds of malignant tumors by extracting meconium with sodium chloride injection. CONSTITUTION:Meconium is treated with sodium chloride injection by applying ultrasonic wave to extract an alkali phosphatase with sodium chloride injection. The alkali phosphatase is precipitated with 60% dry ice acetone, further dissolved in tris buffer solution and dialyzed for 24 hours. Then the dialyzate is purified to give a fraction of a novel alkali phosphatase (meconium ALP for short). The meconium ALP thus obtained shows electrophoresis pattern different from large intestine type ALP, liver type ALP, bone type ALP, cancer type ALP, placental type ALP and intestinum tenue type ALP. Consequently, meconium ALP can be mass produced and is possibly used for diagnosing hepatitis, liver cirrhosis, liver cancer, etc.
