摘要： PURPOSE:To produce an anti-human lymphotoxin monoclonal antibody, by synthesizing a hybridoma using an antigen consisting of a peptide containing the amino acid sequence of human lymphotoxin and free from cytotoxicity. CONSTITUTION:A peptide containing the total or a part of the amino acid sequence of human lymphotoxin and free from cytotoxicity is used as an antigen to synthesize a hybridoma capable of producing an anti-human lymphotoxin monoclonal antibody (anti-LTMoAb). The hybridoma is cultured and the objective anti-LTMoAb is recovered from the supernatant liquid of the cultured product by a proper method. As an alternative method, the hybridoma is transplanted into the abdominal cavity of a mouse preliminarily subjected to pristane-treatment to effect the proliferation of the hybridoma and the anti- LTMoAb is recovered from the ascites of the mouse.

CONSTITUTION:Human lymphotoxin (abbreviated as LT) is purified by hydrophobic chromatography. Concretely, an LT-producing cell is cultured in a serum- containing medium, the obtained cultured liquid is subjected to partial purification to obtain an LT specimen, the LT specimen containing 20% ammonium sulfate is adsorbed to octyl sepharose used as a carrier for hydrophobic chromatography and the adsorbed component is eluted with a buffer solution containing 30% ethylene glycol.

摘要： PURPOSE:To provide an antiviral agent useful for curing viral infections, esp. infections by HIV. CONSTITUTION:The antiviral agent containing, as active ingredient, LFA-3 or analogue (s) thereof, a cell surface protein, cell-adhesive protein distributed over many organs. The LFA-3 molecule which has been known is made up of, starting from its N-terminal, (1) immunoglobulin-like domain 1 (D1 domain), (2) immunoglobulin-like domain 2 (D2 domain), (3) membrane-penetrating domain rich in hydrophobic amino acids (TM domain) and (4) relatively short cytoplasmic domain (C domain). The human LFA-3 molecule has also been known which has D1 domain and D2 domain and is bound to membrane through glycosyl phosphatidylinositol. The human LFA-3 or analogues thereof can be obtained by solubilizing human-derived cells or human erythrocytes using an enzyme or surfactant followed by purification by an affinity chromatography using anti-LFA-3 antibody.

摘要： PURPOSE:To produce an improved factor having long durability in blood, strong affinity to fibrin and decreased side effects by oxidizing a natural type tissue plasminogen activation factor with an active halogen reagent such as chloramine T. CONSTITUTION:A tissue plasminogen activation factor (abbreviated as TPA) produced by the tissue culture of human melanoma, etc., is dissolved in e.g. tris-buffer solution. The obtained solution is adjusted to a weakly alkaline state of pH7-9. An active halogen reagent such as chloramine T is added directly or in the form of a proper solution to the above TPA solution in an amount of >=5 equivalent based on the TPA to start the oxidation reaction. The modified TPA produced by the above process is purified by a method to keep the physiological activity of TPA, e.g. dialysis or ultrafiltration. The obtained factor has improved durability in blood while keeping strong affinity to thrombus and can be used as a remedy for thrombosis such as myocardial infarction and pulmonary embolism and the treatment method attempted at present can be improved by the use of the factor.

An LFA-3 derivative protein not having a D2 region and a TM region, partially having an intracytoplasmic region, and having only a cysteine residue at the carboxyl terminal is immobilized on the prepared carrier through the thiol group of the protein, and CD2 positive cells such as T-cells are subjected to an adsorptive separation to obtain the objective member useful for the prevention of autoimmunity diseases, post-transfusion side effects, T-cell vaccine therapy, etc.
