    • PROBLEM TO BE SOLVED: To provide a gene substitution vector for substituting gene of birds in order to realize a system for producing a foreign protein in yolks of birds including domestic fowl and to provide a method for utilizing the gene substitution vector. SOLUTION: The gene substitution vector comprises a gene encoding a part of vitellogenin protein derived from domestic fowl containing a cathepsin D recognition region. The gene encoding an exogenous Enhanced Green Fluorescent Protein (EGFP) is inserted into the cathepsin D recognition region so as to have a base sequence encoding the cathepsin D recognition region at 3' terminal and 5' terminal of the gene. EGFP having the cathepsin D recognition region at N and C terminals is produced as a vitellogenin fused protein by inducing EGFP gene into domestic fowl hepatocyte (LMH) by using the gene substitution vector.     • PROBLEM TO BE SOLVED: To obtain a vector for producing an objective protein in an egg (especially egg yolk) of a bird including a chicken, to provide a method for producing an objective protein using the vector and a kit containing the vector. SOLUTION: The vector (one example) comprises a promoter and a polynucleotide having a gene encoding a cathepsin D recognition domain, bonded to the 3' terminal side of a gene encoding a domain containing at least a signal sequence and an SE fragment in a chicken-derived vitellogenin protein in which the polynucleotide is controllably bonded to the promoter. The vector has a polynucleotide having a gene encoding a histidine tag, bonded to the 3' terminal side of a gene encoding a cathepsin D recognition domain.
