摘要： PURPOSE: To enable simple and accurate identification of bacteria by a method wherein an antibody being specific to an affinitive bacteria cell surface layer antigen is brought into contact with a bacteria cell to be identified. CONSTITUTION: A reference stock of a bacillus cell, for instance, is cultured, collected, washed and then frozen and dried, and the stocik thus treated is injected into an animal so that this be immunized against it, thereby the reference stock of the bacillus cell is prepared. This stock is injected for immunization into an animal to be immunized, such as a rabbit, rise in an antibody value is confirmed and exsanguination is conducted, thereby blood containing an antibody being specific to a bacillus bacteria cell surface layer antigen is obtained. By using an antiserum containing the antibody obtained from this blood by centrifugation or the like, an antigen-antibody reaction of a bacillus bacteria to be identified with the antibody by an enzyme immunoassay using a labeled antibody is measured. The bacteria to be identified is dispensed into and immobilized in respective wells of a 96 microplate, for instance, and the antiserum containing the antibody is added thereto for reaction. Then, after a secondary antibody being labeled is added, if a coloring agent is added and absorbance is measured by a spectrophotometer, the reaction of the bacteria to be identified with the antibody can be measured simply and accurately.

Action; hydrolyzing raw starch to form oligosaccharide, maltose and glucose. The optimum pH: 6.0+ or -0.5. The optimum temperature: 55 deg.C. Being subjected to nonantagonistic inhibition with S-AI amylase inhibitor, activated and stabilized with Ca . Molecular weight: 78,000 (by SDS-PAGE method). On the other hand, a bacterium capable of producing the enzyme has mycological properties of forming a spore, having flagellum, mobility, 35-45 deg.C optimum temperature and decomposing casein but not DNA.

The obtained strain is subsequently cultivated in a culture medium for highly producing proteins to select the aimed microorganism.

摘要： PROBLEM TO BE SOLVED: To quickly and accurately identify bacteria based on, the amplification of a specific DNA, as indicator by applying PCR method to a chromosomal DNA extracted from bacteria to be identified by using a DNA with specific base sequences present in the 16S ribosome RNA of the bacteria as a primer. The base sequences, specific to Brevibacillus, Aneurinobacillus, Paenibacillus and Thermophilic-Bacillus, respectively, are expressed by respective formulas, I, II, III and IV. The chromosomal DNA extracted from bacteria to be identified is amplified by PCR method, and using the resultant DNA amplified, the genus or group (cluster) of the target bacteria is identified quickly and accurately.
