摘要： PURPOSE:To obtain a recombinant vaccinia virus in high efficiency with a simple operation, by inoculating a mixture of a specific mutant vaccinia virus and a recombinant vaccinia virus to a thymidine kinase-negative animal cell and culturing the cell. CONSTITUTION:A thymidine kinase-negative animal cell is inoculated with a mixture of (A) an attenuated Lister temperature-sensitive mutant vaccinia virus having a proliferation-inhibiting temperature of 40.5 deg.C in rabbit renal cell and giving a pox size of =5 days. The recombinant vaccinia virus (B) is selected from the formed plaque by the plaque hybridization technique using an extraneous DNA.

摘要： PURPOSE:The titled vaccine strain, obtained by attenuating a Lister strain of vaccinia viruses by cell cultivation, and having almost the same antibody production ability as that of the conventional smallpox strain with very weak side reactions of the vaccination. CONSTITUTION:The Lister strain of a parent strain is subcultured in rabbit renal cells at 30 deg.C through 36 passages, and plaque purification is carried out 3 times to separate 50 clones. Temperature-sensitive variant strains, established from renal cells of a green monkey at 40 deg.C, having the worst growth for Vero cells are selected from the 50 clones. The resultant variant strains are then subcultured in rabbit renal cells through 6 passages, and plaque purification is carried out twice to separate uniform clones having relatively small pocks on a hatched chicken egg allanto chorion. The resultant clones are then subcultured in rabbit renal cells at 30 deg.C through 3 passages to separate clones having very small pocks on the hatched chicken egg allantochorion. Thus the aimed attenuated variola vaccine strain is obtained without recognized invasiveness on the central nervous system by peripheral infection of mice.
