    • PROBLEM TO BE SOLVED: To provide an improved method for determining the sequence of a polynucleotide on a single strand fragment level.SOLUTION: The method for determining the sequence of a polynucleotide contributes to the detection of a conformational change in an enzyme that interacts with and processes along the polynucleotide. The detection of the conformational change is carried out by measuring changes in a fluorophore bound to the enzyme. In one embodiment, the method for determining the sequence of the polynucleotide includes a reaction of the target polynucleotide with the enzyme interacting with and processing along the polynucleotide under sufficient requirements for inducing enzymatic activity, and detection of the conformational change in the enzyme in the course of processing the enzyme along the polynucleotide.
