摘要： PURPOSE:To stabilize a carrier in which glutamic acid decarboxylase is fixed for a long duration and provide excellent reproducibility by freeze-drying the carrier in the presence of a solution of a glutamic acid decarboxylase stabilizing agent. CONSTITUTION:Glutamic acid decarboxylase(GAD) may be one which reacts with GAD existing in the blood of a human being and derived from pancreas cells by antigen-antibody reaction and the GAD derived from a mammal is used. An enzyme immunoassay and a particle coagulation reaction can be applied to fix GAD in a carrier. A carrier in which GAD is fixed is, for example, treated for blocking by albumin, serum of a caw, etc. Monosaccharide, e.g. sorbitol, etc., disaccharide, e.g. trehalose, etc., are used for a glutamic acid decarboxylasc stabilizing agent. The stabilizing agent is added to the carrier in which GAD is fixed in an aqueous solution condition. Then the GAD carrier is freeze-dried together with the stabilizing agent solution.

摘要： PURPOSE:To provide a detection process for RNA virus suitable for automation. CONSTITUTION:In a single-pot manner, the sample serum is introduced into a reaction vessel containing the particles to which a single strand DNA 3 having a base sequence complementary to a specific part of RNA 2 in the target RNA virus is immobilized to effect the RNA extraction reaction and immobilize the freed RNA by the hybridization reaction. After the vessel is rinsed, a solution containing transcriptase, a heat-resistant DNA polymerase and an amplification marker labeled with a fluorescent pigment 8 is added to the vessel, whose the temperature is kept at a prescribed temperature to effect the synthesis of cDNA 6 and the amplification of single-stranded DNA labeled with a fluorescent pigment. After the unreacting primer is excluded, a single-stranded DNA is isolated from the particles 11, transferred into a cell for the fluorescent detection 12 to measure the fluorescence to determine the amplified DNA, namely the target virus.

摘要： PURPOSE:To produce a specific antibody which does not react with the VII-th factor of coagulant but react with the VII-th a factor specifically by dosing an animal with tridecaptptide having a specified structure as an antigen thereby immunizing the animal. CONSTITUTION:Tridecapeptide represented by Cys-Leu-Glu-Lys-Arg-Asn-Ala-Ser- Lys-Pro-Gln-Gly-Arg is suspended, as an antigen, in a buffer or a physiological saline and given to a mammal through an interperitoneal or intermuscular route. An antibody having sufficient titer is normally obtained by injecting the antigen three times or more. The blood is sampled upon the lapse of 7 to 10 days after injection and the blood serum thereof is condensed and purified to produce a VII-th a factor specific antibody. Consequently, the VII-th a factor in the blood components, which does not react with the VII-th factor but reacts only with the VII-th a factor specifically, can be determined and the coagulation sthenic state can be determined accurately.

SOLUTION: This hemoglobin A1c composition comprises a hemoglobulin A1c and a sacchrose and is obtained by distributing the whole human blood of a healthy human into centrifugal precipitation tubes, treating the whole blood with a centrifugal separator, removing blood plasmas, blood platelets and leukocytes in the supernatant, separating erythrocytes, adding a physiological saline solution thereto, washing the erythrocytes, adding ice-cooled and purified water to the resultant washed erythrocytes, vigorously shaking the prepared mixture, hemolyzing the erythrocytes, affording a hemolyzate, then passing the purified hemolyzate through a column filled with a support for an affinity chromatogram immobilizing m-aminophenylboronic acid as a ligand, capturing the hemoglobulin A1c , eluting the resultant hemoglobulin A1c and blending the obtained solution of the hemoglobulin-A1c with a stabilizer comprising a 50% aqueous solution of a saccharose. The resultant hemoglobulin A1c composition is stable for a long period and useful for the diagnosis, etc., of diabetes.

CONSTITUTION:There is provided a reagent for examination of virus, containing an oligo nucleotide having a base sequence at least partly composed of a common base sequence derived from a gene coding the same kind of protein in plural different kinds of virus genomes. The common base sequence is derived from a gene coding DNA polymerase in the plural different kinds of virus genomes and one or more of the plural different kinds of viruses are herpes simplex virus 1 type, 2 type, human cytomegalovirus, Epstein-Barr virus or varicella-zoster virus. A pair of primers each having a base sequence common to plural kinds of Herpes viruses and forming elongation reaction products mutually different in its length are used to amplify DNA and the products are detected. The number of times of reaction operations required for the examination can be reduced.
