摘要： PURPOSE:To obtain a biocompatible material having thrombolytic ability by immobilizing a plasmin onto porous crystallized glass consisting essentially of calcium phosphate and titanium by a covalent bond. CONSTITUTION:This material is obtained by immobilizing a plasmin such as urokinase or a tissue plasminogen activation factor onto a porous crystallized glass consisting essentially of calcium phosphate and titanium and obtained from calcium oxide, titanium dioxide, diphosphoruspentaoxide and disodium oxide by covalent bond. The material is a material for thrombolysis, excellent in biocompatibility and having a function capable of continuously converting a plasminogen in blood into a plasmin and suitable for treating thrombosis. The material can also be used as an apparatus for in vivo treatment, stably having thrombolytic action and suitable for decomposing and removing a thrombosis or thrombosis precursor by granulating this biocompatible material and charging the treated material into blood.

摘要： PURPOSE:To obtain the subject complex capable of keeping the activity of a plasminogen activator(PA) effect and effective as a thrombolytic agent for the treatment of cerebral thrombosis, etc., by bonding PA to a porous material composed of a calcium phosphate through covalent bond. CONSTITUTION:A toluene solution of aminopropyltriethoxysilane, etc., is added to a porous carrier (beta-TCP carrier) composed of a calcium phosphate (e.g. beta-tricalcium phosphate) and heated at 110 deg.C for 6hr under refluxing. The treated beta-TCP carrier is washed with toluene and dried. The obtained beta-TCP carrier having introduced amino groups is filled in a column and a 0.1M phosphoric acid buffer solution (pH7) containing glutaraldehyde is circulated through the column. Thereafter, a 0.1M phosphoric acid buffer solution containing a plasminogen activator (e.g. urokinase) is circulated through the column to bond the plasminogen activator to the carrier through covalent bond and the unreacted materials are removed by washing to obtain the objective complex of the plasminogen activator and the porous carrier.

The ratio of the calcium metaphosphate to the tetracalcium phosphate is 1.4-1.75 expressed in terms of atomic ratio (Ca/P) and the burning temperature is 900-1350 deg.C. Apatite is produced on the surface of the objective ceramics by the aforementioned means and tricalcium phosphate is also deposited. The ceramics which are a substrate can firmly be coated with the product by using the calcium metaphosphate.
