摘要： Methods, compositions, and kits are provided for inducing an immune response in an individual (e.g., an individual having cancer). Aspects of the methods include administering an antibody composition having an allogeneic IgG antibody; and administering a treatment that activates antigen presenting cells. In some cases, the antibody composition includes polyclonal allogeneic IgG antibodies with a plurality of binding specificities. In some cases, the polyclonal antibodies are from sera pooled from 2 or more individuals. In some cases, the methods include administering an antigen presenting cell stimulatory agent. Aspects of the methods also include contacting an antigen presenting cell (dendritic cell (DC)) from an individual with a target antigen and an antibody composition having an allogeneic IgG antibody to produce a loaded APC, which can be used to induce an immune response in the individual. Aspects of the methods also include contacting a T cell of the individual with the loaded APC.

摘要： The invention encompasses the discovery that Fc-containing polypeptides that include branched glycans and that are sialylated on the branched glycan (e.g., on an α 1,3 and/or α 1,6 arm in the Fc region's N-linked glycosylation site), with, e.g., a NeuAc-α 2,6-Gal or NeuAc-α 2,3-Gal terminal linkage, are useful in treating neurodegeneration, e.g., in the treatment of neurodegenerative diseases such as Alzheimer's Disease. The present disclosure provides, in part, methods for treating neurodegeneration or neurodegenerative diseases by administering compositions containing such Fc-containing polypeptides as well as methods for evaluating, identifying, and/or producing (e.g., manufacturing) such polypeptides for the treatment of neurodegeneration.

摘要： The present invention discloses a reliable and efficient method of generating an antibody which can discriminate between the membrane bound form of an antigen and the soluble form of the same antigen. Said method comprises the steps of: (i) immunizing an animal with a first antigen which comprises either the membrane bound form or the soluble form of the antigen; (ii) administering to the animal an agent which selectively kills rapidly dividing cells; (iii) when the first antigen of step (i) comprises the soluble form, immunizing the animal with a second antigen which comprises the membrane bound form of the antigen, or when the first antigen of step (i) comprises the membrane bound form, immunizing the animal with a second antigen which comprises the soluble form of the antigen; and (iv) screening for an antibody which can bind to the second antigen but which does not bind to the first antigen.

摘要： The present invention is based on the discovery of polyclonal IgG's ability to promote Schwann cell maturation, differentiation, and myelin production. Methods for treating non-idiopathic, demyelinating peripheral neuropathies in mammals, where the neuropathy is not immune-mediated or infection-mediated, through the administration of polyclonal IgG are provided. Types of demyelinating peripheral neuropathies treatable with the present invention include peripheral nerve trauma and toxin-induced peripheral neuropathies. Alternatively, a composition of polyclonal IgGs can be applied directly to a peripheral nerve cell to induce maturation, differentiation into a myelinating state, and myelin expression or promote cell survival.
