摘要： The present invention relates to adjuvant compositions which are suitable to be used in vaccines. In particular, the adjuvant compositions of the present invention comprises a saponin and an immunostimulatory oligonucleotide, optionally with a carrier. Also provided by the present invention are vaccines comprising the adjuvants of the present invention and an antigen. Further provided are methods of manufacture of the adjuvants and vaccines of the present invention and their use as medicaments. Methods of treating an individual susceptible to or suffering from a disease by the administration of the vaccines of the present invention are also provided.

摘要： The present invention relates to an immunogenic composition having an improved capacity for immuno-stimulation of the immune system of a host to whom it is administered. The present invention especially discloses a composition comprising, as active principle, molecules which would not, as such, elicit the immune system of a host, in a manner sufficient to derive a significant immune response to said active principle and especially to derive a response enabling the use of said composition as a vaccine composition. It relates more particularly to an immunogenic composition comprising a determined active principle and sucrose acetate isobutyrate (SAIB). These immunogenic compositions can also comprise solvents and additives such as bio-degradable polymers or adjuvants . The invention is especially useful for the preparation of immunogenic compositions wherein the active principle is a purified antigen, especially a synthetic or a recombinant antigen or polypeptide against which an immune response in a host is sought.

摘要： A chimeric, carboxy-terminal truncated hepatitis B virus nucleocapsid protein (HBc) is disclosed that is engineered for both enhanced stability of self-assembled particles and the display of an immunogenic B cell or T cell epitope, or both, such as a B cell epitope polypeptide of the influenza M2 protein and a T cell epitope of the influenza NP protein. An immunogenic epitope is peptide-bonded to one or more of the N-terminus, in the immunogenic loop or at the C-terminus of HBc, whereas the enhanced stability of self-assembled particles is obtained by the presence of at least one heterologous cysteine residue near the amino-terminus of the chimer molecule. Methods of making and using the chimers are also disclosed.

摘要： The present invention relates to methods and compositions for augmenting an immunogenicity of an antigen in a mammal, comprising administering said antigen together with an adjuvant composition that includes glycosylceramide, preferably alpha-galactosylceramide (alpha-GalCer). According to the present invention, the use of glycosylceramide as an adjuvant is attributed at least in part to the enhancement and/or extension of antigen-specific Th1-type responses, in particular, CD8+ T cell responses. The methods and compositions of the present invention can be useful for prophylaxis and treatment of various infectious and neoplastic diseases.

摘要： A method of enhancing protective immunity induced by any polynucleotide vaccine or combination of polynucleotide vaccines comprising immunizing with a priming immunization mixture of a polynucleotide vaccine mixed with a polynucleotide vector that expresses a cytokine. Subsequently the priming vaccination is followed by immunizing with a boosting immunization preparation that contains a recombinant pox virus. This method also reduces the amount of vaccine needed to achieve protective immunity and broadens the coverage in a vaccinated population.
