摘要： An a-amylase assay method is described together with a reagent kit for use therein. An oligosaccharide substrate for a-amylase is provided. The substrate contains at least 3 glucose units. Its reducing-end glucose unit is bonded, via a bond cleavable by a- or β- glucosidase, to a label which exhibits an optically measurable change upon cleavage of the bond. Its terminal glucose unit is bonded to a blocking substituent which inhibits cleavage by exo-enzymes of the bond between the terminal glucose unit and the adjacent glucose unit. A liquid assay sample is contacted with the oligosaccharide substrate and with an exo-enzyme capable of cleaving the bond between the reducing-end glucose unit and the label. The optically measurable change is used as a measure of a-amylase in the sample.

摘要： Provided herein is a method for the quantitative screening of hydrolase for desired substrate activity using pH indicators which are sensitive to the release of protons from a chemical reaction in a reaction mixture. The method comprises selecting buffer and indicator conditions such that both have the same affinity for protons such that the relative amount of buffer protonated is proportional to the amount of indicator protonated as the pH of the reaction mixture shifts. A reaction mixture is then prepared comprising a buffer, indicator, hydrolase to be tested, and desired substrate to be tested, allowing the hydrolase to react with the substrate. The reaction is monitored by detection of change in color of the reaction mixture, determined by the affect of the reaction on the indicator.
