    • PROBLEM TO BE SOLVED: To provide a nucleic acid amplification reaction including amplification using a polymerase chain reaction, and an assay method using the reaction and obtained by combining a sequence method and a hybridization probe detection method.SOLUTION: There is provided a method for producing both a double-stranded chain amplicon and a single-stranded chain amplicon by extending an oligonucleotide primer with a DNA polymerase, and analyzing at least one single-stranded chain amplification product in an asymmetry nucleic acid amplification step including at least one primer annealing temperature. The method includes: detecting the two-stranded chain amplicon by detecting homogeneous fluorescence; detecting the at least one single-stranded chain amplicon product by detecting the homogeneous fluorescence sequence-specific in a lower temperature than the at least one primer annealing temperature; and calculating a fluorescence ratio of the single-stranded chain product to fluorescence of the double-stranded chain amplicon.
