The CD1d complexes comprise a CD1d, a ß2-microglobulin molecule, and may further comprise an antigen bound to the CD1d binding groove. The invention is further directed to methods of inhibiting or stimulating an immune response with the CD1d-antibody compounds, in particular anti-tumor and autoimmunity responses.

摘要： The invention is directed to methods of modulating an immune response in an animal, comprising administering a composition comprising one or more soluble CDId complexes, in particular non-specific soluble CDId complexes. Soluble CDId complexes comprise a soluble CDId polypeptide, a β2-microglobulin polypeptide, and a ceramide-like glycolipid antigen bound to the CDId antigen binding groove, and in certain embodiments, an immunogen. The administration of compositions of the present invention affects the activity of CD Id-restricted NKT cells, and in particular, allows for multiple administrations without causing CD Id-restricted NKT cell anergy.

摘要： Compositions and methods are provided for treating diseases associated with CDlOO, including certain types of cancers, autoimmune diseases, inflammatory diseases including central nervous system (CNS) and peripheral nervous system (PNS) inflammatory diseases, transplant rejections, and invasive angiogenesis. Compositions include anti-CD100 antibodies capable of binding to a soluble human CD1OO antigen or a CD1OO antigen located on the surface of a human CD1OO- expressing cell, wherein the antibody has CD1OO blocking activity that is achieved by having at least one optimized CDR or FWR engineered within the variable region of the antibody. Compositions also include antigen-binding fragments, variants, and derivatives of the monoclonal antibodies, cell lines producing these antibody compositions, and isolated nucleic acid molecules encoding the amino acid sequences of the antibodies. The invention further includes pharmaceutical compositions comprising the anti-CD100 antibodies of the invention, or antigen-binding fragments, variants, or derivatives thereof, in a pharmaceutically acceptable carrier, and methods of use of these anti-CD100 antibodies.

摘要： The present invention is directed to a novel targeted vaccine delivery system, comprising one or more peptide-MHC Class I complexes linked through the β 2 -microglobulin molecule to an antibody which is specific for a cell surface marker. The complexes of the invention contain a β 2 -microglobulin that has been modified to have greater affinity to the α chain of MHC Class I than native β 2 -microglobulin. Alternatively, the complexes of the invention contain β 2 -microglobulin fused or linked to the antigenic peptide. The complexes of the invention are useful for treating and/or preventing cancer, infectious diseases, autoimmune diseases, and/or allergies.
