    • PURPOSE:To make it possible to inhibit nonspecific adsorption of a labeled substance by bonding polylysine covalently or noncovalently to the surface of an insoluble substance thereby covering the surface. CONSTITUTION:An antigen or an antibody is not bonded directly or indirectly to a carrier but the surface of the carrier is covered with polylysine by bonding the polylysine covalrntly or noncovalently thereto thus inhibiting the surface properties of the carrier itself causing nonspecific bonding. The carrier includes polystyrene, vinyl chloride resin, alumina, metal, for example. The labeled substance includes a radioisotope, an enzyme, a fluorescent substance, for example. When polylysine is bonded to a carrier and blocking is conducted using a protein which does not participate in the immunoreaction, a carrier having lower nonspecific adsorption can be obtained. The protein includes bovine serum albumin gelatin, casein, for example. In other words, nonspecific adsorption can be inhibited by improving the properties on the surface of the carrier.

    • PURPOSE:To make it possible to blocking effect in a short time by treating a carrier for antibody or antigen for a specified time in a solution of phthalic acid and/or citric acid containing a protein and the like which do not participate in the immunoreaction. CONSTITUTION:Carrier for antibody or antigen is polystyrene, PVC resin, alumina, metal, etc., to which an antibody or an antigen is bonded by physical adsorption. Blood serum albumin, gelatin, or casein, for example, is employed as a protein which does not participate in the immunoreaction. Phthalic acid, citric acid or succinic acid having concentration in the range of 20-150mM and pH in the range of 2-4 is employed as an acid solution. The carrier is treated in the acidic solution for about 1 to 30min to cause acidic modification of proteins which do not participate in the immunoreaction and to increase the adsorption rate of the carrier thus realizing efficient blocking.
