    • PURPOSE:To safely immunologically measure phosphatidylcholine (PC) with a simple operation and high sensitivity in a short period of time by using a monoclonal antibody which specifically recognizes the PC. CONSTITUTION:The PC which is prepd. from the egg yolk, is immobilized on nitrocellulose paper and is homogenated in PBS is directly immunized as an antigen to the spleen of a mouse. The PC formed by coating acid treated Salmonella Minnesota bacteria with the PC is additionally immunized from the vein of the tail part after two weeks. The spleen is extracted from the mouse after 3 days of the final immunization and the antibody forming cells are prepd., by which the hybridoma is prepd. The analysis of the supernatant of the hybridoma culture is then executed and the hybridoma producing the antibody which reacts specifically with the PC is selected. Plural kinds of the monoclonal antibodies which specifically recognize the PC are thus obtd. The independent measurement of the PC in a sample by identifying the same from other phospholipids is, therefore, possible. The measurement of the many specimens with the simple operation and the high sensitivity in a short period of time is thus executed.

    • PURPOSE:To widen a linear area when a calibration curve is made by adopting a 2 monoclonal antibody sandwich method using an immobilized 1st antibody and 2nd antibody which is labeled by a labeling agent. CONSTITUTION:The 1st antibody which is the monoclonal antibody to recognize TBG and is immobilized and the 2nd antibody which recognizes the antigen section different from the 1st antibody and is labeled by the labeling agent are used in the method of measuring the human thyroxin binding globulin TBG in a sample by an immunological method. The monoclonal antibody is preferably obtd. by fusing the antibody genic cells of an animal immunized by TBG and myeloma cells to obtain the hybridoma which produces the monoclonal antibody to recognize TBG, then culturing this hybridoma or the cell strains derived therefrom and drawing the monoclonal antibody from the cultured matter thereof. The solid phase immune enzyme immunoassay by the sandwich method using the two antibodies is, therefore, enabled. The conjugation constant of the 1st and 2nd antibodies is preferably 10 -10 l/mol.

    • PURPOSE:To immunologically measure the human pancreas alpha-amylase with high accuracy and high sensitivity in a wide concn. range with a simple operation in a short period of time by using a monoclonal antibody specific to the human pancreas alpha-amylase. CONSTITUTION:The human pancreas alpha-amylase is immunologically measured by a sandwich method using the 1st antibody A immobilized by the monoclonal antibody which specifically recognizes the human pancreas alpha-amylase and does not make cross reaction with the human salivary gland alpha-amylase and the 2nd antibody B which recognizes the antigen section different form the antibody A and is labeled by a labeling agent. The antibodies may be immobilized by any known method. The sample is not restricted at all and any known method may be used for the labeling of the antibodies. There are no restrictions on order of adding the sample, the antibody A and the antibody B in the sandwich measuring method and either method of 1-step sandwich and 2-step sandwich may be used. Washing, etc., of a B/F sepn. method, etc., to be used in ordinary immunoassay may be executed to remove the free labeled antibodies.
