    • 1,153,114. Amino acids; nucleosides by regulated fermentation. AJINOMOTO CO. Inc. 23 Jan., 1968 [6 Feb., 1967], No. 3612/68. Heading C2C. [Also in Divisions C6 and C7] A fermentation process involves culturing micro-organisms in a culture medium in a closed system, removing from the closed system carbon dioxide produced during the process, agitating the culture medium and feeding oxygen to the closed system at a desired controlled rate. A measurement of the partial pressure of oxygen in the liquid or gas phases within the system may be used for on-off control of an electrolytic cell providing oxygen, or of an oxygen cylinder. Alternatively, the amount of oxygen produced by an electrolytic cell 21, Fig. 3, operated under constant voltage potential may be regulated by the variation in level of the electrolyte 21 in the region of the electrode 20 produced by changes in the partial pressure of oxygen in the gas phase of the closed fermenter 1. In this embodiment the oxygen analyser 17 and recorder 18 do not participate in the control action. Carbon dioxide produced is removed by continuously circulating the gaseous phase through a solution of sodium or potassium hydroxide in a washing bottle 6. The fermenter itself may be agitated. Examples relate to the production of L- glutamic acid by cultivation of Corynebacterium acetoacidophilum ATCC 13870 on a specified glucose-containing culture medium in which a dissolved oxygen content of 0À05 atm. at 31.5‹ C. was maintained, as by periodic actuation of a 1 1. capacity electrolytic cell connected to a direct current stabilized power source of 5-15 volts; the production of 5-amino-4-imidazolecarboxamide-riboside by cultivation of Bacillus megaterium ATCC 15117 on a specified culture medium containing glucose and ribonucleic acid (dissolved oxygen=0À02 atm. at 34‹ C.); the production of inosine by cultivation of Bacillus subtilis ATCC 13952 on a specified culture medium containing glucose and adenine (dissolved oxygen=0À06 atm. at 31‹ C.); and the production of L-glutamic acid by cultivation of Corynebacterium hydrocarboclastus ATCC 15110 on specified n-alkane containing culture media (dissolved oxygen partial pressure=0À06 atm. or partial pressure of oxygen in the gas phase= 0À6 atm. at 30‹ C.).
